Objective: Laryngeal carcinoma is a common head and neck malignant tumor and a serious hazard to human health. Multiple genes(e.g,Rb, C-MYC, MDM2, PTEN and other laryngeal carcinoma related genes)and multiple pathways have participated in its occurrence and development.The Rb gene is found the first tumor suppressor gene in humans and mainly plays a suppressor role through regulating the cell cycle. With the further study of Rb gene, we find that Rb gene is closely related to the invasion and metastasis of laryngeal carcinoma, and local recurrence or metastasis of laryngeal carcinoma determines the human morbidity and mortality.According to elaborating the relevance of Rb, C-MYC, MDM2 and PTEN in laryngeal cancer from this paper, we may provide the theoretical basis for Rb on laryngeal cancer pathogenesis and represent novel potential therapeutic strategy for clinical intervention.Method: Using CNKI, VIP, Wan Fang, PubMed retrieval system, to retrieve the literatures(including original works, literature review) about the express of tumor-associated genes such as Rb, C-MYC, MDM2 and PTEN in laryngeal carcinoma during 1999 to 2016, and then tocomprehensive analysis on Rb. Database of search terms is: Laryngeal neoplasms, Retinoblastoma protein gene, C-MYC, MDM2, and PTEN.Results: Many domestic and overseas literatures reported that laryngeal cancer, one of the high incidence of respiratory cancer, is harm to the quality of life and life safety. Pathogenic factors of laryngeal Carcinoma not only involve smoking, alcoholism, laryngopharyngeal reflux and gastroesophageal reflux, also involve tumor suppressor genes and oncogenes in the host cell. With high expression in laryngeal carcinoma,cancer-causing gene C-MYC stimulates glycolysis mainly by controlling the glycolysis mRNA, thus contributing growth and survival of laryngeal cancer cell. There is MDM2 gene amplification in laryngeal carcinoma and The expression of MDM2 may be often associated with tumor grading,pathological staging and tumor metastasis. PTEN participates in maintaining genomic stability and its shortcoming will contribute to laryngeal cancer cell proliferation, reduce apoptosis, and facilitate migration. Rb gene inhibits cell proliferation, stimulates differentiation and apoptosis, participates in DNA replication and repair, metabolism, cell survival, the maintenance of stem cells quiescent period and plays a central role in homeostasis and biological cells. Deletion of Rb gene is associated with laryngeal Dysplasia. Rb, C-MYC, MDM2 and PTEN promote the development of throat cancer through the interaction of multiple paths.Conclusion: Tumorigenesis is a complex process of multiple factorsand multiple stages. Rb tumor suppressor genes may be interacted with C-MYC, MDM2 and PTEN affecting the larynlx occur and development.the specific mechanisms of Rb taking part in cancer development and transition are worthy of further study, making Rb gene and its protein become a target of early diagnosis and treatment of laryngeal cancer.
