Objective: 1.To investigate the expressions of human papillomavirus type 16 E6(HPVl6E6) and p53 in human oral cancer.2.We observe the distribution and expression of TLR1(Toll like receptor, TLR1) in cancerous and adjacent tongue squamous cell carcinoma tissue, and explore the role of TLR1 in the pathogenesis of tongue squamous cell carcinoma.Method: 1.The positive rates of HPV16E6 and p53 were detected by the immunohistochemical method in 149 specimens with oral cancer, 88 specimens with precarcinoma of oral cancer and 49 specimens of normal oral tissues. And the positive rates of HPV16E6 and p53 were detected in different clinical stage and histopathological grade.2. The m RNA and protein levels of TLR1 in 60 patients with tongue squamous cell carcinoma samples were measured using reverse-transcriptase polymerase chain reaction(RT-PCR), western blot. Immunohistochemistry and immunofluorescence were employed to study the characteristic of distribution of TLR1 in cancerous and adjacent tissue.Result: 1.In oral cancer tissues, precarcinoma of oral cancer tissues and normal oral tissues, the positive rates of HPV16E6 were 79.9%(119/149),38.6%(34/88) and 2.0%(1/49), respectively, and that of p53 were 24.2%(36/149), 55.7 %(49/88) and 92.0%(45/49), respectively. Compared with normal oral tissues, the positive rate of HPV16E6 was significantly higher in oral cancer tissues and precarcinoma of oral cancer tissues, and that of p53 was significantly lower in oral cancer tissues and precarcinoma of oral cancer tissues(P<0.05). The positive rate of HPV16E6 in clinical stageⅡ, Ⅲ, Ⅳ were significantly higher than that in clinical stageⅠ, and that of p53 was significantly lower in clinical stageⅠ(P<0.05). The positive rate of HPV16E6 in histopathological grade Ⅱ, Ⅲ were significantly higher than that in histopathological gradeⅠ, and that of p53 was significantly lower in histopathological gradeⅠ(P<0.05)。2 RT-PCR analysis and western blot analysis showed that TLR1 had decreased expression in tongue squamous cell carcinoma specimens than that in matche d adjacent tissues(P<0.01). The expression levels of TLR1 closely related of the clinical parameters of deeper tumor invasion, survival. Immunohistochemistry results showed TLR1 was mainly expressed in tongue normal squamous cell and vascular endothelial cell of duct, and weak expression in squamous cell carcinoma tissues. The immunofluorescence staining further revealed that TLR1 was co-location with the endothelial cell of blood vessels marker CD31, not co-location with the smooth muscle aorta cells mark er α-SAM.Conclusion: 1.Overexpression of HPV16E6 and low-expression of p53 may involved in the genesis and development of human oral cancer. 2. The high expression of TLR1 in tongue carcinoma were significantly decreased closely associated with clinical p rogression, indicating that TLR1 plays a role in the developed process of tongue squamous cell carcinoma; it may be as an important prognostic indicator in clinical judgment.
