Objective: To study the expression of osteopontin (OPN) in oral verrucous carcinoma (OVC) and oral squamous cell carcinoma(OSCC), so as to investigate the role of this gene in the development of OVC and OSCC, and to validate the results of gene chip.Methods: 9 cases of exogenic OVC,6 cases of cystoid and infiltrative OVC,15 cases of well-differentiation OSCC (wd-OSCC) and 15 cases of poor-differentiation OSCC (pd-OSCC) were selected; two specimens, which were carcinoma tissues and the surround normal tissues, were taken from the same patients. Test the mRNA expression of OPN gene by reverse transcription polymerase chain reaction (RT-PCR) and check the OPN protein by immunohistochemistry (IHC) in the above specimens.Results: 1,The OPN results of RT-PCRa,The mRNA expression of OPN gene was significantly higher in both exogenic OVC and cystoid and infiltrative OVC than in their matched normal tissues (P<0.05); The mRNA expression of OPN gene also significantly higher in both wd-OSCC and pd-OSCC than in their matched normal tissues (P<0.05).b,The mRNA expression of OPN gene was significantly higher in cystoid and infiltrative OVC than in exogenic OVC(P<0.05). c,There was no difference in the mRNA expression of OPN gene between wd-OSCC and pd-OSCC(P>0.05).d. The mRNA expression of OPN gene was significantly higher in wd-OSCC and pd-OSCC than in the two types OVC(P<0.05).2,The OPN protein expression by IHCa,Negative or weakly positive expression of OPN was showed in normal mucosa where it major located in the plasm in the positive one. Positive expression of OPN was showed in all exogenic OVC,cystoid and infiltrative OVC,wd-OSCC and pd-OSCC, which the locations of OPN were not only in the plasm of cancer cell, also in epithelia and endothelia cells around the edges of cancer nub.b,The intensity score (Is)of OPN in 2 types OVC and 2 types OSCC was higher than in their matched normal tissues (P<0.05).c,There was no difference of OPN's Is between wd-OSCC and pd-OSCC(P>0.05).d,The Is of OPN was significantly higher in OSCC than in OVC (P<0.05).e,The Is of OPN was significantly higher in this order: the lymph node metastasis oral squamous cell carcinoma(LSCC)>cystoid and infiltrative OVC>the lymph node metastasis oral squamous cell carcinoma(LSCC)>exogenic OVC (P<0.05). Conclusions: 1,OPN may play an key role in the invade and metastasis of OVC and OSCC; and it may be related to the promotion both in the interact cancer cell with matrix and form new tumor veins.2,The difference of OPN in OVC and SCC may be the gene foundation of invade and metastasis that the potential of OVC is lower than OSCC。3,And it also showed part of cystoid/infiltrative OVC can have a higher potential of invade and metastasis than OSCC.4,There is a relationship between OSCC'S clinic stages and lymph node metastasis with OPN, but the tumor's differentiation.
