Safflower(Carthamus tinctorius L.) is a traditional Chinese herbal medicine, which contains variety of active ingredients, and has been widely used in clinical drug research, health care products, food industry and other fields. In this study, 20 groups of safflower samples collected from different areas in Xinjiang were studied. The main research contents were:1. Using the microwave assisted extraction technology with distilled water as extraction agent, the extraction process of effective components of safflower were investigated by taking the single factor test and orthogonal test, the optimal extracting conditions of safflower effective ingredients were : extraction time:5min, extraction temperature :70℃, solid-liquid ratio :1:50(g/ml), microwave power :600W.2. The sample was digested by microwave digestion method, and the optimum microwave digestion conditions were determined. The results showed that the safflower sample and the soil sample were digested completely. ICP-AES method was used to measure the contents of 23 kinds of trace elements in the 20 groups of samples from different habitats. The correlation coefficients of the established method were all over 0.9975, the precision of RSD<2.07%, and the recovery rates were 91.21%~106.3%. This method is sensitive, accurate and can be used to simultaneous determination of trace elements, the content of trace elements and trace heavy metal elements in Chinese herbal medicine and herbal origin soil samples.3. The infrared spectrum of each sample was obtained by using the FTIR spectroscopy technique, 12 common peaks were observed and determined. The obtained dates, processed by using statistical and chemical pattern recognition techniques to compare the similarity, and by taking the principal component analysis and cluster analysis. Data show that the 20 groups of samples can be roughly divided into 3 categories, the difference between regions is quite obvious.4. The content of hydroxyl safflower yellow A in 20 groups of samples from different sources was determined Under the optimized condition. The results showed that there was a significant difference at the hydroxyl safflower yellow A content between different origin, the content of effective components in northern region was higher than that of the southern region. The method has the advantages of short separation time and good separation effect, simple, accurate and reproducible, and can be used for the determination of the content of hydroxy safflower yellow A.5. The five main active components in safflower is separated and analyzed by HPLC-MS. The content of each component in 20 groups of samples were determined by the HPLC. The result shows that The correlation coefficient of the standard curve were all over 0.9998, the established method is accurate and the precision of the instrument is good, and it reached the requirement of the measurement. The data were processed by using statistical techniques and chemical pattern recognition method to take the principal component analysis, cluster analysis, The spectrum and the related data analysis can reflect the intrinsic quality of safflower in a certain extent. It provides the technical support for the quality control, the authenticity identification and the development and utilization of the medicinal herbs.
