Tussilago farfara is the only species that belongs to Tussilago genu. The buds of Tussilago farfara is called Kuandonghua in Chinese it is an important and well known Chinese traditional drug. It is used in the treatment of cough, bronchitis, and asthmatic disorders. Modern pharmacological studies have shown that Tussilago farfara L. has the bio-activities of antitumor, hypoglycemic, antioxidant, anti-inflammatory, relieving cough, phlegm, asthma, anti platelet activating factor, promoting bile secretion, etc. This thesis deals with the investigation of chemical constituents and antibacterial and antioxidant activity of Tussilago farfara. and secondary metabolites of endophytic fungi from Tussilago farfara.The phytochemical constituents of Tussilago farfara L. were studied by using variety of modern chromatographic separation methods(chromatography on silica gel, etc.) and spectro-scopic identification analysis(EI-MS, IR, et al). 25 compounds were isolated and purified, the structure of 18 compounds were elucidated: 2-Aminoethyl tetradecanoate(1), 24-methylenecycloartane-3β,22-diol(2), β-amyrin-n-nonyl ether(3), Tussfarfarin B(4), Isobauerenol(5), 2α, 3β-dihydroxylurs-12-en-28-oic acid(6),(4S)-3, 4-dihydro-4, 8-dihydroxy-l(2H)-naphthalenoe(7), Methyl 2α-hydroxyursolate(8), 7β-[3’-ethyl-crotonoyloxy]-lα[2’-methylbutyryloxy]-3.14-dehydro-Z-notonipet-ranone(9), β-sitosterol(10), Ursolic acid(11), Ixocarpalactone B(12), 3,4-di-O-caffeoyl quinic acid methyl ester(13), Moluccanic acid methyl ester(14), 4,5-di-O-caffeoylquinic acid butyl ester(15), 2-(3’-O-β-D-Glucopyranosyl-4’-hydroxyphenyl)-ethanol(16), S(-)-2-(2-hydroxypropanamido) benzamide(17), Chlorogenic acid(18).Reducing power and DPPH radical and hydroxyl radical scavenging ability were tested to evaluate the antioxidant activity of polarity fractions extracts from Tussilago farfara. The results showed that antioxidant activity of ethyl acetate extract is better than vitamin C. Ethyl acetate extract and N-butanol extract also have obvious antioxidant activities.In addition, the antibacterial activity of all the extracts was determined by Oxford cup method. The results showed that ethyl acetate extract has the best antibacterial activity. Pe-troleum ether extract has higher inhibition against Staphylococcus aureus and Escherichia coli. N-butanol extract have higher inhibition against Staphylococcus aureus.In addition, this thesis study antioxidant activity and antibacterial activity of secondary metabolites of endophytic fungi and mycelium methanol extracts from Tussilago farfara. The results showed that the plant secondary metabolites of four endophytic fungi have stronger inhibitory effect on five tested bacteria. Antioxidant activity of the secondary metabolites extracted from ethyl acetate in the fermentation medium may reach about 50% of the Vc activity. Explanation, the chemical composition has good biological activity of bioactive secondary metabolites.The purpose is make it haspossible find new natural active ingredients from endophy- tic fungi of Tussilago farfara.The thesis reviews phytochemical and pharmacological activity research progress of the genus Tussilago and describes the phytochemical research,antibacterial and antioxidant activity of Tussilago farfara. This research provides a scientific basis for the further develop-ment and utilization of Tussilago farfara L. medicinal plant resources.
