Objective: the euphorbia sororia schrenk has been included in the 44(3)issuance of Flora Reipublicae Popularis Sinicae, which only states its name, character, origin place, source, and identification method. However, Fascicule of Uyghur Medicine issued by the Ministry of Health of People’s Republic of China has records about the character, identification method, impurity, moisture check, function, and indication of fructus euphorbiae sororiae. Application quality standards and requirements of the euphorbia sororia schrenk have not been established. Through scientific research about chemical components and quality control of the euphorbia sororia schrenk, this paper could promote better development and utilization of the medical material with Xinjiang Uyghur characteristics.Method: types of chemical components in the euphorbia sororia schrenk can be judged on basis of the systematic preliminary test for chemical components. This paper measures the moisture, extract, total ash, acid-insoluble substance of 10 batches of the euphorbia sororia schrenk. According to requirements of Chinese Pharmacopoeia(2015 edition), this paper uses ICP-AES to measure Fe, Cu, Zn and Cr, employs GFAAS(graphite furnace atomic absorption spectrometry) to measure Pb and Cd, and adopts AFS(atomic fluorescence spectrometry) to measure contents of As and Hg. Contents of total polyphenol and general flavone can be measured by using ultraviolet and visible spectrophotometer. In addition, this paper explores rules of infrared spectrum of the euphorbia sororia schrenk and its extracts by measuring10 batches of euphorbia sororia schrenk through infrared spectrum and chemometrics analysis. This paper also carried out qualitative research about three effective ingredients of 10 batches of euphorbia sororia schrenk including kaempferol, scopoletin, and vanillic acid through HPTLC(high performance thin layer chromatography). Moreover, contents of these three effective ingredients in the euphorbia sororia schrenk can be measured through HPLC(high performance liquid chromatography).Resu Lts:the outcome of the systematic preliminary test for chemical components shows chemical components of the euphorbia sororia schrenk mainly include flavonoid, phenol, saccharides, and alkaloid etc. Average values of the moisture, total ash, water-soluble extract, and ethanol-soluble extract in 10 batches of euphorbia sororia schrenk are respectively 5.74%, 11.53%, 17.68%, and 14.82%.After measuring contents of 8 designated elements(Fe, Cu, Zn, Cr, Pb, Cd, As, Cr)through ICP-AES, GFAAS, and AFS, it can be found the 8 designated elements comply with requirements of Chinese Pharmacopoeia(2015 edition) for element contents. By optimizing technologies of extracting total polyphenol and general flavone from extracting solution of 10 batches of euphorbia sororia schrenk, it is found the optimum extracting technologies and conditions for total polyphenol and general flavone depend on an ethanol concentration of 70%, a solid-liquid ratio of1:40, an ultrasonic temperature of 45℃, and an ultrasonic time of 30 minutes. In addition, their contents shall be respectively maintained at 9.89-14.33 and18.01-23.14 mg/g. The 10 batches of euphorbia sororia schrenk vary from each other in peak numbers, peak positions, peak forms, and peak intensities of infrared absorption peaks. By comparing infrared spectrums of samples, this paper finds the peaks of 3391cm-1, 2927cm-1, 1324cm-1, and 1240cm-1 could be taken as common characteristic peaks of euphorbia sororia schrenk. This paper classifies the 10 batches of euphorbia sororia schrenk into 3 types through PCA(principal componentanalysis) and clustering analysis. Kaempferol, scopoletin, and vanillic acid in the batches of euphorbia sororia schrenk present better effect in separation of thin layers. This could be taken as an indicator for quality evaluation of euphorbia sororia schrenk. This paper uses HPLC to measure contents of kaempferol, scopoletin, and vanillic acid in euphorbia sororia schrenk, so as to offer theoretical bases for medical use of euphorbia sororia schrenk. Conclusion:this paper aims to identify a content measurement method of kaempferol, scopoletin, and vanillic acid in euphorbia sororia schrenk through preliminary exploration about the quality evaluation method of the Uyghur medicine euphorbia sororia schrenk. In addition, this paper offers scientific bases for evaluation and research of the Uyghur medicine euphorbia sororia schrenk by measuring contents of different elements in euphorbia sororia schrenk.
