The thesis adopt Ermiao Wan,Sanmiao Wan,Simiao Wan and Jiawei Simiao Wan as basic and categorized formula on the basis of TCM theory.According to main pathological feature of anti-damp-heat syndrome gout,We choosed anti-inflamation,analgesia, decreasing serum uric and immune study as effective index,in order to evaluate the material foundation and biological effct of anti-gout from the whole formula to effective parts.Meanwhile,we did some research on compatible mechanism.Then,dig the bioactive effective parts and constituent and study their compatible mechanism.We study the logical correlation according to hierarchical cluster analysis,and explore the correlation of material foundation and biological effect on anti-damp-heat-syndrome gout of Ermiao Wan categorized formula.In the end,we explore the compatible rule from basic to categorized formula.The main research content as follow:First of all,we adopt mouse acetic-acid-induce twisting experiment,mouse xylene-induced auricular swelling experiment,an animal model of hyperuricemia to decreasing serum uric acid and carbon particle clearance test to evaluate the anti-damp-heat-syndrome gout pharmacodynamic effect of Ermiao Wan categorized formula and categorized formula without polysaccharide,and evaluate the difference of effective regularity of Ermiao Wan categorized formula.Then,analysis pharmacodynamic difference of Ermiao Wan categorized formula and categorized formula without polysaccharide and definite material foundation of immune function.And then,we prepared the samples of effective parts of Ermiao Wan categorized formula.We adopt mouse acetic-acid-induce twisting experiment,mouse xylene-induced auricμlar swelling experiment,an animal model of hyperuricemia to decreasing serum uric acid and carbon particle clearance test to evaluate the pharmacodynamic effect of effective parts,and make sure the bioactive parts.After that,we study ICAM-1 expression in HUVEC induced by H2O2 from cell level.Then,we study the anti-inflamation mechanism of anti-damp-heat-gout of Ermiao Wan effective parts and analysis the relationship of pharmacodynamic effect. The more important research is to make compatible mechanism of effective parts clear.Separation,purification,content determination were studied to establish the pharmacodynamic foundation for further study.We adopt mouse carbon particle clearance test,cellular and humoral immunity experiments to approach the polysaccharide part’ effect mechanism of nonspecific immunity and specific immunity.Then,Volatile oil,total saponin,total alkaloid,total flavonoides,organic acid and sugar were separated for the research of decreasing serum uric acid experiment according to orthogonal combination. Through the experiment,we can get compatible mechanism of decreasing serum uric. After that,we set up the human vascuar endothelial cell(HUVEC) damaged model induced by urate to search for the activity of HUVEC induced by urate and study the compatible mechanism of effective parts which control the activity of HUVEC.Further,we approach the influence on the activity of effective constituent which control the HUVEC through setting up the HUVEC damaged model induced by urate and H2O2.In the end,We analyze biological effect on anti-damp-heat-syndrome gout and Ermiao Wan categorized formula and effective parts through hierarchical cluster analysis,in orther to explore the correlation of material foundation and biological effect on anti-damp-heat-syndrome gout of Ermiao Wan categorized formula.
