Study On The Hemostatic Mechanism And Quality Standard Of Carbonized Rhizoma Imperatae

Rhizoma imperatae,is the dry rhizome of Imperata Cylindrica. This dissertation basicly studies on the hemostatic mechanism and quality standard of carboned Imperata Cylindrica. Through pharmacodynamics research, we demonstrated the ethyl acetate extract and tannin as the chemicial effective part or index composition of carboned Imperata Cylindrica. Based on basicly studies on the technology of processing of Imperata Cylindrica, we have done technology of semi-works processing of Imperata Cylindrica. Combined with phamacodynamics results, tannin content was used as a quality standard control index of carboned Imperata Cylindrica.The dissertation is composed of four parts, the literature summary and the research, technology of semi-works processing of Imperata Cylindrica, hemostatic mechanism study, quality standard study,and fingerprint study. The main research contents and results were as follows:1. The literature summary:1.1 Study on the history of processing of Imperata cylindrica:Study on the history of processing of Imperata cylindrical and previous original intention. Summary previous processing experience and clinical experience and provide useful reference for developping study on standardization of TCM decoction pieces.1.2The modern literature summary: evaluate and analysis on chemical, pharmacological and clinical application of Imperata cylindrica and make strict basis for making feasible studying scheme.2. Research progress on hemostatic mechanism of Charcoal of Chinese drugsConsult and classify the documents of traditional Chinese medicinal herbs from ancient time to now.With reference to hemostatic mechanism of Charcoal of Chinese drugs, ancient presented "using black to win red,the bleeding stop meeting with black" based on five elements theory;Modern research taxonomic study on material basis of hemostatic mechanism of Charcoal of Chinese drugs. And hemostatic mechanism of Charcoal of Chinese drugs is very complex, that is not effect on a single factor, but that complex effects on multiple link and target.3. Study on pilot precessThrough orthogonal design,the technology parameters were described as 280℃,4min.4. Study on hemostatic mechanism4.1 Pharmacodynamic studyBy means of pharmacodynamics research, we demonstrated the ethyl acetate extract and tannin as the chemicial effective part or index composition of carboned Imperata Cylindrica.4.2 Study on tannin content in Imperata Cylindrica before and after processingStudy on tannin content in Imperata Cylindrica before and after processing, tannin contents of semi-works productions have 155.50% degrees accrescence.That provides a basis for futher hemostatic mechanism study,because tannin is a kind of hemostais active composition.4.2 Study on adsorbability in Imperata Cylindrica before and after processingStudy on tannin content in Imperata Cylindrica before and after processing, carbon element contents of semi-works productions are 102.1% of before processing .That provides a basis for futher hemostatic mechanism study.5. Study on quality standardAccording to Chinese pharmacopoeia loaded assay method, we had done moisture determination of ten batch of pilot product .And regulate the moisture determination of them were not less than 5%. We had done water-soluble extracts determination of ten batch of pilot product. And regulate the water-soluble extracts determination of them were not less than 5%.Combined with phamacodynamics results, tannin content was used as a quality standard control index of carboned Imperata Cylindrica.As result of producting area and harvest time difference, We sadatame temporarily the control index of carboned Imperata Cylindrica were not less than 0.2%.6. Study on fingerprint by HPLC of pilot precess pieces for total quality control of carboned Imperata Cylindrica.
