Objective:In order to obtain the preparation of antibacterial and antitussive effects of Tibetan medicine, simultaneously provide a new thinking for curing oral cavity ulcer, Tibetan medieinal Plants Dracocephalum heterophyllu were collected from Qinghai Province and that of antibacterial were studied in vitro and in vivo. Antitussive and curing oral cavity ulcer activity were also investigated. The preparation of total extracts were investigated and discussed.Method:1. The antibacterial activity of D. heterophyllu in vitro:Use the agar diffusion method and with inhibition zone for the evaluation index to assess the antibacterial activities of total extracts and essential oil in vitro.2. The antibacterial activity of total extracts in vivo:Mice peritonitis model were established by infected with Staphylococcus aureus and Pseudomonas aeruginosa respectively to valuate the antibacterial efficacy of D. heterophyllum in vivo.3. The antitussive activity of total extracts:Mice cough model were induced by stronger ammonia water, and Pentoxyverine Citrate Tablet was as positive control to valuate the antitussive efficacy of D. heterophyllu.4. The preparation of total extracts:Mixed total extracts and excipients, and then wet granulating.5. The essential oil relative content of D. heterophyllu was detected by gas chromatography-mass spectrometry.6. The essential oil curing oral cavity ulcer activity:Rats oral cavity ulcer model were established by infected with Staphylococcus aureus and Candida albicans respectively to valuate the essential oil of D. heterophyllum on curing oral cavity ulcer efficacy.Results:1. The total extracts and essential oil of D. heterophyllum possess moderate antibacterial activity against tested bacterial stains. The inhibition zones were in the range of 13-15 mm and 20-21 mm, respectively.2. The total extracts of D. heterophyllum may be out of invalid in vivo.3. The result of antitussive efficacy:the total extracts of D. heterophyllum could also decrease the frequency of cough induced by ammonia in mice and prolong the latent period of cough.4. Prepared a standard Capsules of D. heterophyllum.5. GC-MS analysis resulted in the identification of thirty-seven compounds of essential oil D. heterophyllum, which consisted mainly of epoxy compound, alcohols, esters, ketones, aromatic compounds and derivants, etc.. 6. As compared with the positive control group, essential oil D. heterophyllum provides similar efficacy to positive control group in the improvement of oral cavity ulcer.
