Objective:This research focused in the activation of muscle skeletal satelittle cell and the dynamic change of the activation factors(Myo D, HGF, MGFm RNA) in Kingming mice during detraining.We analyzed the dynamic changing features of the activation factors and explored possibility mechanism of skeletal muscle disuse atrophy of detraining.The results provided the reference information for the treatment and the proventment of muscular dystrophy by detraining.Methods:48 Healthy five-week-old male KM mice were randomly distributed into control group(C group,n=8) and experimental groups(E group, n=8) after adapting environment for 3 days, and training treadmill for 3 days. Then, the experimental groups were randomly divided into groups of E0(0h, n=8), E1(12h, n=8), E2(24h, n=8), E3(3d, n=8), E4(7d, n=8) after stopping running. C group rats were fed commonly for 8 weeks, while experimental groups were assigned an 8-week training program increasing load by a certain degree. At the last running, Left and right hind leg quadriceps were measured from the mice, whose neck was taken off. Using immunohistochemical, Elisa, RT-PCR test method for measuring the activation of muscle skeletal cell and the dynamic change of the activation factors(Myo D,HGF,MGFm RNA). After 8-week running, E1 group、E2 group、E3 group、E4 group’s wasn’t suspened until 12h、24h、3d、7d respectively. The statistical results of all data was representation of average value ± standard deviation( x ±SD). C, E0, E1, E2, E3,E4 group were analyzed by One-Way ANOVA, significant difference for p<0.05, very significant difference for p<0.01.Results:All indexs rised in the first stage, and then decrease during distraining.(1)Total wet weight from quadriceps reached maximum at 24 h,and very significantly increased(p<0.01);(2)The activate number of SC reached maximum at 12 h,and very significantly increased(p < 0.01);(3)The expression of Myo D reached the peak number at 24 h,and very significantly increased(p<0.01);(4) The expression of HGF reached the peak number at 24 h,and very significantly increased(p<0.01);(5) The expression of MGFm RNA reached the peak number at 12 h,and very significantly increased(p<0.01).Conclusion:(1)From wet weight analysis of muscle in mice,having been reached the maximum at 24 h detraining,after began to decline;(2)From the perspective of activation of SC, the activation of SC reached the maximum at 12 h of detraining,and has been fully restored at 7d of detraining;(3)Each index was similar at different time of detraining,and reached the maximum during 12-24 h of detraining.12-24 h was the best moment to prevent muscle atrophy.
