The termite gut is a relatively closed microenvironment,the symbiotic microorganisms in the gut of termites also have their unique properties.They can help the termite break down the lignocelluloses which is difficult to digest and produce the stimulation of the growth of termites,etc.This study was based on the diversity of microorganisms in the gut of termites.The study by high throughput sequencing technology and pure culture method in order to research the diversity of bacteria in the gut of termite and screening strains with lignocellulose decomposing ability and antibacterial activity.The termite gut bacteria 16 SrDNA gene V4 region was sequenced by high-throughput sequencing.Finally get 62404 effective sequences,630 OTUS.In total 13 phyla of bacteria,including Firmicutes,Bacteroidetes,Proteobacteria,Actinobacteria,Spirochaetes.Among them,the Actinomyces accounted for 16.95%.In this study,the microbes in the gut of termite were separated by different culture media and isolation methods.A total of 1458 strains were obtained,including actinomycetes 327 strains.The 5 suborders of actinomycetales,inciude Streptomycineae,Micrococcineae,Corynebacterineae,Propionibacterineae,Micromonosporineae.A total of 10 families and 18 genera.And a new classification is separated.YIM 110061 was identified as a new genus by preliminary analysis.In this study,the strains isolated from the strain were screened for measurement of cellulase activity and lignin enzyme activity.The curve of cellulase activity with time was measured in four strains,The cellulase activity of YIM 110167 was the best.In this study,three kinds of lignin enzyme activities were measured,including laccase,peroxidase,manganese peroxidase.The peroxidase activity of YIM 110063 was the highest,the activity of the other two enzymes was also detected.The filter paper was screened,it was found that the filter paper was broken and the liquid culture medium became turbid five days later.In this study,327 strains of actinomycetes were screened for antimicrobial activity and functional genes.The 11 strains inhibited Escherichia coli,22 strains inhibited Bacillus subtilis,30 strains inhibited Staphylococcus aureus,32 strains inhibited Streptococcus white,48 strains inhibited Aspergillus.Detection of six functional genes of 327 strains of Actinomycetes.The termite gut is a very special environment,there are a large number of strains to be studied.
