Studies On Anatomical Characteristics And Phylogenetic Analysis Of Tomeutella In Inner Mongolia

The genus Tomentella forms abundant ectomycorrhizae in coniferous and deciduous,so its has very important significance in forest ecosystems.Although Tomentella has a widely distribution,its basidiocarp is easily neglected because of its resupinate character,is therefore rarely found.The research of Tomentella in China is relatively scare.In this study,specimen were collected from Helan Mountain National Nature Reserve,Saihanwula National Nature Reserve,Heilihe National Nature Reserve,Baiyinaobao National Nature Reserve,Gen He city,Great Khingan,used a combination of molecular-phylogenetic inference of ITS rDNA sequence and anatomical approach to ananlyzed the classification of Tomentella in Inner Mongolia.The results of this study are as follows:1.The macroscopic and microscopic morphology of the samples collected from 5 collection sites were observed,and detailed described based on the color and morphological characteristics of the hymenophore and subiculum,the color,size and the characteristics of subicular hyphae,subhymenial hyphae and rhizomprphs,the shape,size and characteristics of cystidia,badia and basidiospores and the ornamentation of basidiospores to devide the samples into 21 different types.The color of hymenophore and subiculum of the basidiocarp in Inner Mongaolia is yellow brown to black brown,and there were two kinds of surface characteristics of the hymenophore;some species has rhizomprphs,the color of rhizomprphs is colorless to black,with 4 kinds of characteristics.The color of subicular hyphae is range from colorless to dark brown,and including 4 kinds of characteristics.The color of subhymeninal hyphae is colorless to dark brown,it has 3 kinds of characteristics.Small part of the basidiocarp with a cystidia,the color is colorless to brown,with 2 shapes.The color of basidia is colorless to dark brown,with 2 shapes and 7 kinds of features.Basidiospores color is deep yellow to dark brown,and there was 4 kinds of shapes and 2 types of ornamentation.2.Molecular-phylogenetic analysis of ITS rDNA secquence of 21 samples has indicated that all the species belongs to Tomentella,there were 8 species forming a independent clade with the sequence of Tomentella from Genebank,and the posterior probabilities is 0.95-1.Combined with morphological comparison,we found that 2 of them were new record species in China which were Tomentella fibrosa and T.terrestris,respectively.Other samples may contain new species of Tomentella,due to the scarcity of reference materials in China at present,so it needs further study.
