Permafrost is widely distributed in the global and permafrost ecosystem is a huge carbon pool and its stability has a significant influence on the global environment and climate change.Microbial species are well contained in the permafrost regions and play an important role in the element circulation,mineral formation and transformation of the permafrost.These microorganisms possess unique structural and physiological characteristics communities.In November 2008,Samples of gas hydrate were collected in Qilian Mountain Permafrost DK-1 drill.which is the gas hydrate samples were first discovered in the high altitude permafrost zone of low latitudes.Surface soil samples sediments of seven line,DT,DZ2(2011),DT,DH,DS,DZ2 and DZ3(2012),surrounding permafrost zone of Qilian Mountains drill,were analyzed for water chemistry parameters,total organic carbon(TOC),CH4,acridine orange direct count(AODC)and microbial diversity.The conclustions are as following:The microbial abundance in surface soil were 108 cells/g or above,and which higher in surface soil than subsurface samples.Methanosarcinales was the most abundant fraction in sample from 2011,which accounted for 30%of all samples.Group I.3b was the dominant group in sample from 2012.which accounted for 31%of all samples.Archaea were less of study so far,the function of many groups was not clear;community structure of sample from drilling regional and reference area have obvious difference.Especially methanogens,it is distributed more widely than the 2012samples.The reasons for this phenomenon is unknown.Conform to the characteristics of the methanogens anaerobic,the content on subsurface sample is significantly higher than in the surface sample.It's no significantly correlation between environmental factors and most of microbial groups.Cluster analysis shows that archaea community structure has significant differences under the different ecological environment.In this research,we study enrich the database of frozen soil archaea community.These results provided new perspective for frozen soil microorganisms and environmental research.
