In recent years,microfluidic technology as a new interdisciplinary field,in the life sciences cell detection,electrochemical measurement,microelectronic integrated circuit structures and other fields have great potential.In this paper,based on the principle of dielectric micro-droplet control technology and cell impedance measurement technology,to achieve the micro-droplet transport and electrochemical impedance measurement functions such as microfluidic chip design,set up a digital microfluidic system,To achieve the integration of the chip,miniaturization and portable goals.Mainly include the following aspects:Firstly,based on the analysis of cell impedance measurement technique,an interdigital array microelectrode for impedance measurement of two-electrode system was designed to improve the rate and sensitivity of reactants in cell impedance measurement.The cell 2D culture on microelectrode And cell impedance miniaturization measurements.The influence of the electrode size and the material on the impedance spectroscopy was analyzed and simulated.An equivalent circuit model of the adherent cell layer was proposed,and the equivalent circuit components were analyzed and explained.The cell layer existed Cytoplasmic resistance and cell membrane capacitance.Second,determine the control of the digital microdroplets by dielectric wetting.The effects of dielectric wetting effect,driving electrode structure,dielectric layer material and thickness,hydrophobic layer material and thickness on the potential distribution of digital microfluidic system were analyzed.The selection and preparation of the impedance measuring electrode,the driving electrode,the dielectric layer and the hydrophobic layer are completed.Finally,the test platform of the digital microfluidic chip system and the design and preparation of the digital micro-droplet drive circuit were constructed.The transport of 0.5 ?L microdroplets was carried out on the 40 V voltage.The MDCK cells,HUVEC cells and Hep G2 cells were The impedance measurement and dynamic measurement of impedance spectroscopy were used to analyze the impedance spectra of Nyquist and Bode diagram,equivalent circuit component fitting parameters and return to standard treatment.The results showed that the resistance value and membrane capacitance value of MDCK cell layer,HUVEC cell layer,And Hep G2 cell layer cell death process impedance changes.
