Cells are widely present in living organisms and marine environments.The detection and analysis of cells will help advance the development of life sciences,disease diagnosis,and marine pollution identification.The detection of intrinsic biomarkers,size and impedance properties of cells is one of the important research directions.The current general equipment for detecting these cell parameters is mainly large instruments such as flow cytometry and fluorescence microscopy.Which has the disadvantages of large volume,complicated operation,high price,and inability to perform rapid analysis on site.Based on this,this paper studied the cell detection technology based on microfluidic chip,and tested the intrinsic biomarkers,size and impedance characteristics of the cells.(1)Based on the fluorescence characteristics of cells,the laser-induced fluorescence detection technology was studied and a fluorescence detection system was designed and built in this paper.A microfluidic chip with a fluorescence detection structure is used as a detection platform.The cells are excited by the light source to generate fluorescence,the mechanical components are used to fix the system components,the fluorescence receiving module is used to receive the fluorescent signals of the cells,and the filtering amplification circuit is used to process and collect the fluorescent information.The fluorescently labeled cells are identified and counted based on the magnitude and number of amplitudes of the resulting fluorescent pulses.(2)Based on the size characteristics of cells,the DC impedance pulse sensing technology was studied,and a DC impedance pulse sensing system was designed and built in this paper.A microfluidic chip with a DC impedance detecting structure is used as a detection platform.The mechanical components are used to fix the system components,the DC impedance information of the cell is sensed by the electrode,and the filter amplifier circuit is designed to process and collect the DC impedance information.Cells are identified and counted based on the magnitude and number of amplitudes of the resulting DC impedance pulses.(3)Based on the different characteristics of various cells on the resistance and phase of the detection channel,the AC impedance pulse sensing detection technology was designed and constructed in this paper.A microfluidic chip with an AC impedance detecting structure is used as a detection platform.The mechanical components are used to fix the system components,the AC impedance information of the cell is sensed by the electrode.Phase-locked module based on AD630,the forward and reverse envelope detection module,and the filter amplification module are used to process and collect the AC impedance information.The cells are identified and counted based on the magnitude and direction of the resulting AC impedance pulse.(4)To verify the feasibility of these three detection systems in this paper,experiments were performed using four samples of nasopharyngitis cancer cell CNE1,lung cancer cell H1299,Pyramidomonas sp.and polystyrene particles with a size of 15?m.The experimental results show that cells can be detected and classified based on the differences in cell characteristics according to the detection technique proposed in this paper.In addition,the newly developed equipment has the advantages of small size,low cost,simple operation and low cost.
