Polyrhachis vicina Roger was a social insect belonging to Insecta,Hymenoptera,Formicidae.It had a direct or indirect benefit relation with Human life.In recent years,with Polyrhachis vicina Roger medicine,pharmacology,clinical research,people paid more and more attention to the medicinal value and health care resources development and utilization.Polyrhachis vicina Roger was used for characteristics of traditional Chinese medicine(TCM)without the analysis of the nutrients to the ant,analysis of various kinds of active physical substance,didn't make full use of the ant contains rich protein,crude fat and mineral elements and formic acid and other nutrients.In this paper,Northeast Polyrhachis vicina Roger was used as materials provided by Leixin insect food factory Xiaoxian Anhui Province.Mainly studied the basic component of Polyrhachis vicina Roger including the protein content,fatty content,formic acid content,the main mineral content,type and content of amino acids;The optimum conditions of ultrasonic-assisted extraction of Polyrhachis vicina Roger fat was studied,and the fatty acids in Polyrhachis vicina Roger were esterified by two methods and analyzed by GC-MS,the components and relative contents of the fatty acids esterified by two methods were compared;The hydrolysis parameters were optimized by Response surface methodology(RSM);Polyrhachis vicina Roger enzymolysis liquid were blended by traditional Chinese medicine extract such as Gynostemma pentaphyllum Makina,honeystuckle,licorice,selfheal.These had practical help and significance for the whole of the development and application of ant products.The main research results were as follows:1.Polyrhachis vicina Roger protein content reached 62.7%,especially the content of essential amino acids,it was a high-quality protein source.It was rich in mineral elements,especially for Fe,Zn content in Polyrhachis vicina Roger reached430.67mg/100 g and 336.7mg/100g;Formic acid content is 35.35mg/g.2.The optimum conditions of ultrasonic-assisted extraction of Polyrhachis vicina Roger fat were studied,and the fatty acids compositions were esterified by two methods and analyzed by GC-MS.By the single factor and orthogonal tests,the factors of affecting the extraction rate of Polyrhachis vicina Roger fat were studied,such as the granularity of raw materials,ultrasonic power,temperature,ultrasonic time,and fatty acids were analyzed by GC-MS.The results indicated that the optimum extraction were as follows: the granularity of raw materials 60 meshes,ultrasonic power 100 W,temperature 50?,ultrasonic time 100min;In these conditions,the extraction rate of Ant fat was 27.5%.The predominant fatty acids of Polyrhachis vicina Roger fat were octadecenoic acid,palmitic acid and stearic acid by two esterification methods.The octadecenoic acid and palmitic acid content,respectively,71.307% and 11.096% were identified by the second esterification method.3.Effect of Alkaline Protease on enzymolysis of Polyrhachis vicina Roger was studied for the subsequent deployment of drinks.The amount of each single factor was determined by single-factor experiments,on this basis three extraction parameters including pH,hydrolysis time,enzyme substrate ratio,the experiments according to the Box-Behnken of RSM were designed,Content of peptides as response.The optimum conditions were extracted pH 9.4,enzymolysis time 5.3h,enzyme substrate ratio(%)1.0.Under this condition,the R value 20.20mg/mL,the fit of the model prediction is really good.This revealed the applicability of RSM to the optimization of Alkaline Protease enzymolysis process of Polyrhachis vicina Roger protein4.Polyrhachis vicina Roger and Gynostemma pentaphyllum Makina as main raw materials coupled with honeystuckle,licorice,selfheal were used to prepare a kind of compound health beverage,Gynostemma pentaphyllum Makina and The Ant enzymolysis liquid(7%)between the mixing ratio of 1:4 were determined by single experiment,then The Polyrhachis vicina Roger Gynostemma pentaphyllum Makina compound fluid and honeystuckle,licorice,selfheal were studied by means of orthogonal rest design.Results showed that the optimal conditions for The Polyrhachis vicina Roger Gynostemma pentaphyllum Makina compound fluid and honeystuckle extract,selfheal extract,licorice extract of compound beverage for10:3:2:2.The compound beverage was 0.30 NTU by ceramic membrane filtration,sugar and citric acid were added to get the best taste of compound beverage.The compound beverage storage test under three different temperature conditions such as refrigerate 4 ? and room temperature,constant temperature of 37 ?.The results showed that the three kinds of temperature storage test pH value and soluble solids content kept stable,turbidity and L value at 37 ? and room temperature changed greatly,but under the condition of 4 ? refrigerated storage changed much,recommended storage at low storage at low temperature,which is beneficial to keep drinks the quality of the original.
