| Monosodium glutamate(MSG)and Sodium nitrite(NaNO2)as two of the food additives are widely used in our lives,and whether MSG is harmful to human health has been debate in many reports.In order to investigate the effects of these two food additives in animals,the fed results of MSG(7.5mg/ml)and NaNO2(0.72mg/ml)for the different caeste adults of Polyrhachis vicina Roger(P.vicina)were detected in death rate,weight change and shape change aspects.Beside that some important genes of P.vicina including ecdysteroid receptor(EcR),ultraspiracle protein(USP),cyclic AMP response element binding protein(CREB),Estrogen related receptor(ERR)and glutamine synthetase(GInS)genes were detected.RNA interference experiments were performed on the PvEcR and PvCREB genes to confirm the degree of gene influence of these two food additives.The above research results were offer as the follows:1.Compared with the control group,the mortality of workers,females and males was significantly higher after fed MSG solution.the females' weight increased,and appearance appeared deformity,such as,the abdomen of a few P.vicina including the workers,females and males were distended and deformed.2.In the MSG experiment,the PvEcR,PvUSP,PvCREB,PvERR,PvGInS mRNA expressions were down-regulated detected by qRT-PCR technique,among which the workers were the most significant.3.Compared with the control group,the mortality of workers,females and males increased significantly after fed NaNO2 solution.The appearance of these P.vicina no obvious changes.4.In the NaNO2 experiment,PvEcR,PvUSP,PvCREB,PvERR,PvGInS mRNA expressions were down-regulated in different degrees detected by qRT-PCR technique,among which the males were the most significant.5.In the PvEcR-RNAi experiment,compared with the control group,the mortality of P.vicina were increased after fed PvEcR-dsRNA solution,among which workers and males were the most significant,The appearance of workers and females have no obvious change,a few males abdomen appeared deformity.There was no significant change in the workers 'body weight,while females increased weight on days 3,9,12 and males lost weight on days 3 and 9.After PvEcR-RNAi,compared with the control group,the PvCREB mRNA expressions of all adult were down-regulated,while the workers of the PvUSP mRNA expressions were down-regulated,and the females and males's expressions shown a trend of fluctuation,such as down-regulated,or up-regulated and down-regulated detected by qRT-PCR technique.6.In the PvCREB RNAi experiment,compared with the control group,the mortality rates of the workers,females and males were increased after fed PvCREB-dsRNA solution,among which workers and males were the most significant.The appearance of workers,males have no obvious change,a few the females abdomen appeared deformity.There were no significant change in body weight of all the P.vicina.After PvCREB-RNAi,compared with the control group,the PvUSP mRNA expressions of,the females,workers and males were down-regulated,while PvEcR mRNA expressions of these P.vicina were shown a trend of fluctuation,such as down regulated,up-regulated and down-regulate detected by qRT-PCR technique.The results show that MSG and NaNO2 were harmful for the development of adults of different castes of P.vicina,which may be related to disturb normal expression of PvEcR,PvUSP,PvCREB,PvERR,PvGInS genes in the adults of P.vicina.This study also confirmed that the effect of MSG and NaNO2 as food additives were similar to results of interfered PvEcR and PvCREB by RNAi for the adults of P.vinica.The present study also indicated that the PvEcR,PvUSP and PvCREB genes have important regulatory roles for the development of different castes adult ants of P.vicina.Moreover,PvEcR,PvUSP and PvCREB may have some correlations at the mRNA level.Finally,for the sake of health,it is recommended that people eat little or no food containing monosodium glutamate and sodium nitrite food additives,and advocate moderate intake. |
