The rice as raw material and daqu as saccharifying leaven used by lamb rice wine, This wine fermentation process add fresh mutton soup,Fresh fruits such as pear and rare Chinese traditional medicine type. Lamb rice wine's saccharifying is daqu, daqu in our country is often as saccharifying leaven of distillation type liquor, So this wine without distillationg process has a unique quality by use daqu as saccharifying leaven. There are a variety of microbes and enzyme system in daqu which is the main power in the winemaking process, has the "the bone of wine " said. There has been no reports of microbes diversity in the lamb wine's qu. The topic of this paper main research the lamb of microbial diversity in daqu wine, which could contain a variety of microbes including yeast, mold and bacteria. Show the microbial diversity of daqu to improve wine quality and reveal the formation mechanism of liquor flavor and the quality of the wine has the vital significance.In order to obtain the different organisms of this daqu,The subject through direct separation,augment cultivation, fermentation cultivation. Three ways to treat the microorganism in daqu.at last, Author isolated 528 strains of yeast, mold isolated a total of 102 strains, isolated 80 strains of lactic acid bacteria,the other bacteria were isolated 715 strains.After through to the colony morphology observation and microbial cells observation, according to the differences of different microbial morphology has carried on the cluster analysis, the yeast poly mold for 14 kinds of morphological types, divided into 12 types of bacterial morphological types, divided into 15 types of form. Finally,by molecular methods to 14 different morphological types of yeast identify to 6 types of molecules, and sequencing of the 6 types of yeast, appraisal results:Pichia anomala,Saccharomyces cerevisiae,Trichosporon asahii,Issatchenkia orientalis, Cryptococcus albidus,Rhodotorula mucilaginosa. saccharomyces cerevisiae distinction into subspecies level, seven kinds of different forms of saccharomyces cerevisiae is divided into five classes the subspecies level.Establish fungal DNA extraction method, author examining four fungal DNA extraction methods, enzymatic hydrolysis, thermal cracking method, microwave extraction, freezing mortar method, finally established the frozen mortar for fungal DNA extraction. Using molecular techniques to the distinction between two similar mould into molecular level, and finally the two mould identification for the same kind of mold. After the colony morphology observation and cell morphology observation for identifying different types of mold, the appraisal results are:Penicillium corymbiferum,Rhizopus peka,Aspergillus oryzae,Penicillium adametzi,Aspergillus niger,Absidia corymbifera.In the separation of various microbes, From the number and frequency can be obtained, Saccharomyces cerevisiae is the main yeast in the daqu, and Absidia corymbifera is the major fungal flora in the daqu.
