Seabuckthorn (Hippohae rhamnoides L). department of elaeagnaceae hippophae is a natural medicinal and edible plant. Seabuckthorn resources are widely distributed in Europe and Asia. In our country, it is mainly distributed in the northern, northwest, southwest region. Seabuckthorn roots, stems, leaves, flowers and fruit have rich nutrients and bioactive substances. In this paper, seabuckthorn seeds was used as raw material, and removed grease with petroleum ether and flavonoids and procyanidins with ethanol, then prepared seabuckthorn seed protein with alkali extraction and acid precipitation.The effects of hypoglycemic, anti-inflammatory and gene expression of seabuckthorn seed protein on diabetic mice were studied. The efficacy and mechanism to provid theoretical basis were explored for the development and utilization of seabuckthorn seed protein products.Seabuckthorn seed protein content and yield were determined by protein automatic tester, amino acid composition was determined by automatic amino acid analyzer. Then mice were fasted for 12h before administration of STZ, buffered in cold sodium citrate by intraperitoneal injection. Mice model of type 2 diabetes were lavaged treatment 4 weeks with seabuckthorn seed protein, seabuckthorn polysaccharides and seabuckthorn procyanidins. We research seabuckthorn seed protein in diabetic mice on body weight (BW), fasting blood glucose (FBG), blood lipid, serum insulin (SIN) and insulin resistance. We used ELISA to determined serum c-reactive protein (CRP), tumor necrosis factor-a (TNF-a), a nuclear factor-?B (NF-?B), interleukin-6 (IL-6), AMP-activated protein kinase (AMPK) and Silent information regulator 1 (SIRT1). Using the method of semi-quantitative PCR (RT-PCR) to study the effect of seabuckthorn seed protein on gene expression in diabetic mice liver, such as glucose-6-phosphatase (G-6-P), glycogen synthase kinase-3 (GSK-3), carnitine palmacy transferase 1-a (CPT1-a) and glycogen phosphorylase (GP).The results showed that seabuckthorn seed protein content was 70.1%, the yield was 36.5%, and amino acids full range and essential amino acid uniform composition. Animal experiment results showed that groups of seabuckthom seed protein, seabuckthom polysaccharide and seabuckthom procyanidins treatment fasting blood glucose (FBG), total triglycerides (TG), total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C), serum insulin level and insulin resistance index (IRI) were significantly lower than model group (p<0.05), high density lipoprotein cholesterol (HDL-C), insulin sensitivity index (ISI) were significantly higher than that of model group (p<0.05). Groups of seabuckthorn seed protein, seabuckthorn polysaccharide and seabuckthorn procyanidins treatment insulin inflammatory cytokines in mice were significantly lower than that of model group (p<0.05), hepatic AMPK, SIRT1 activity in mice were significantly higher than model group (p<0.05). According to the results of RT-PCR groups of seabuckthorn seed protein, seabuckthorn polysaccharide and seabuckthorn procyanidins treatment were significantly lower than model group on G-6-P, GSK-3, CPT1-a and GP mRNA expression levels.The results of this study showed that SSP has a significant hypoglycemic and anti-inflammatory effect as evidenced by the lower BW, FBG levels, SIN and lipid contents of diabetic mice treated with SSP compared to the diabetic control mice. ELISA experiments showed that seabuckthorn seed protein can lower insulin resistance and inflammatory factor levels in mice of type 2 diabetes, and increased efficiency of AMPK, SIRT1 activity. RT-PCR results showed that seabuckthorn seed protein can inhibit gene expression of G-6-P, GSK-3, CPT1-a and GP. Its mechanism may be related to enhanced AMPK, SIRT1 activity and inhibit gene expression of G-6-P, GSK-3, CPT1-a and GP. Seabuckthorn seed protein can effectively prevent and control the occurrence of type 2 diabetes.
