Study On The Hypoglycemic Activity Of Effect Of Nanfengmi Citrus Vinegar And Its Mechanism

Diabetes is a group of metabolic diseases characterized by hyperglycemia caused by absolute or relative insulin deficiency.It is another serious disease that endangers human health after tumor,cardiovascular and cerebrovascular diseases.Nanfeng honey orange is a unique variety of honey orange in Jiangxi province.It has great significance to develop it into honey orange vinegar.By constructing a STZ induced diabetic rat model,the hypoglycemic effect of Nanfeng honey orange vinegar was investigated and its hypoglycemic mechanism was preliminarily discussed.Normal blank group of SD rats(normal saline gavage),control group(according to 0.8 ml/kg/d Nanfeng honey orange fruit vinegar gavage),diabetes model control group(model after gavage normal saline),low and high dose model group(model after gavage 0.2 ml/kg/d?0.8ml/kg/d Nanfeng honey orange fruit vinegar),positive control group(model after gavage apocarbose)and apple vinegar raw vinegar group(model after gavage 0.8 ml/kg/d apple vinegar raw vinegar),conducted a comparative study of blood sugar reduction.effect of nafeng honey orange fruit vinegar(0.8 ml/kg/d)was not significantly different from that of the positive control group(apoca sugar).Based on ?-inhibition of glucoxycase activity,regulation of hepatic glucose metabolism and antioxidation,the mechanism of hypoglycemic effect of Nanfeng honey orange fruit vinegar was studied.The results show that:(1)Inhibition of ?-glucosidase activity.The inhibition rate of glucosidase in ?-was 42.6% and 58.2%(p<0.05),and there was no significant difference between the high concentration fruit vinegar group and the positive control group.Compared with the inhibition rate of glucosidase in the small intestine mucosa ?-rats SD Nanfeng fruit vinegar group,apple vinegar group and diabetes model group,it was found that Nanfeng fruit vinegar could effectively inhibit ?-glucosidase activity.(2)Regulation of liver glucose metabolism by Nanfeng honey orange fruit vinegar.Intraperitoneal injection of alanine solution showed that high dose Nanfeng honey orange fruit vinegar group could effectively reduce the conversion rate of gluconeogenesis(49.50%)and promote the synthesis of liver glycogen.The content of liver glycogen in rats treated with Nanfeng honey orange fruit vinegar group was 6.43 mg/;.(3)Antioxidant Effect of Nanfeng Citrus VinegarDPPH radical scavenging rate,hydroxyl radical scavenging rate and superoxide anion scavenging rate were 52.31%,52.99% and 62.57%,respectively.The levels of superoxide dismutase(SOD),catalase(CAT)and reduced glutathione(GSH)in the liver homogenates of SD rats were measured to compare the antioxidant effects in the treatment groups.The results showed that high dose of Nanfeng honey orange vinegar could effectively enhance the enzyme activity of SOD and CAT.The above studies show that Nanfeng honey orange fruit vinegar has obvious hypoglycemic effect,and its possible mechanism includes slowing down intestinal carbohydrate absorption and regulating liver sugar metabolism.
