Chemical glycosylation of biomaterial surface is essential for mimicking the dense "glycocalyx" of biomembrane and the development of bio-functional materials. In this study, a convenient method was developed to conjugate carbohydrates onto different surface using divinyl sulfone (DVS) as the coupling.Mannose and lactose glycosylated films were prepared and the bioactivities were assessed at protein and cell level. The protein level results showed that the glycosylated films can specifically recognize Concanavalin A (Con A) and Peanut agglutinin(PNA), respectively. The cell adhesion experiment using macrophages (RAW 264.7) and HepG2 suggested that the glycosylated film is highly bioactive, low cytotoxic and suitable for medium and long-term cell culture in vitro. The histidine modified film was also prepared, it can specifically adsorb immunoglobulin G(IgG), showing great potential on antibody purification.The vinyl sulfone coupled lactosamine was synthesised used for the modification of surface containing amino group. The results showed that the functionalized surface can specific adsorb PNA. HepG2 could also attached on the surface and retain good biological activity, showing great potential for the large scale cultivation of hepatocyte.A versatile, stable and facile method was developed for the immobilization of carbohydrates on the surface containing hydroxyl or amino group. The simplicity of this method would have significant impact on the construction biomimetic membrane and glycomics research.
