Nanotechnology is a kind of applied science in order to study the design,composition,characteristics and application of nano-materials.The fast growing number of applications of nano-materials in biomedicine,new energy based transportation,defence construction and so on.This means that human exposure to nano-materials is expected to increase greatly.According to data,nano-particles will accumulate in the kidney through circulation in the body.There is obvious increase in the risk of kidney damage.And the gold nanoparticles always are chosen as model to study the biological effect.Here,we prepared different size of gold nanoparticles(10 nm,25 nm,50 nm,70 nm,100 nm)to explore the damage caused by them on subcellular organelles and the size dependent.We found that gold nanoparticles can cause golgi stress through the change of lysosomal membrane permeability and the leaching of Ca2+ of ER.The protein glycosylation was blocked and the cell adhesion was lower.In this study,we have uncovered a damage on organelle that influence protein to synthesis and secrete normally by treatment of cells with 50 nm Au NPs.This study can improve the biological mechanism between nanoparticles and cells.It is also meaningful for the cytotoxicology,application and development of nanoparticles.
