The Separation And Purification Of Moringa Seed Extract And Its Appication For Coagulation Removal Of Dyes From Water

Experiments were carried out to extract the proteins from Moringa oleifera seed presscake?MOSP?,and then the standard jar tests were conducted to investigate the dyes wastewater removal using the proteins as coagulant.The results indicated that the optimal extracting process was as follows: the mixture of MOSP and 0.5mol/L?NH₄?₂SO₄ solution at the rate of 1:20?W:V?was treated with an ultrasonic cell disrupter for 5 minutes.The mixture was filtered and?NH₄?₂SO₄powder was added into filtrate until the filtrates was 80% saturated by?NH₄?₂SO₄,then the solution was centrifuged at 4000 rpm for 15 min to obtain the white precipitate,the precipitate was finally dialyzed in the dialysis tube overnight to get the purified proteins.The collected liquid was further purified using the protein separation and purification system.The active component of the purified liquid was water-soluble proteins which can remove dye from water by coagulation.Its molecular weight was about 19 KDa determined by using protein gel electrophoresis analysis.The standard jar tests were carried out the test the coagulating behaviors of the dialyzed protein for the removal of four direct dyes,including Direct Fast Black G,Direct Yellow R,Direct Light Turquoise Blue GL and Direct Fast Yellow 5gl.PAC at the same dosage was used to carried out the same experiments as comparison.The results indicated that the best removal condition was: setting time=90 min,temperature=30 ?,pH=8.The removal of the four dyes increased with the increase of protein dosage.Compared with the PAC,the proteins was better for the removal of the four dyes in all the experiments.This part of the research work is only started work.In the follow-up study.We willcontinue to explore the optimization of moringa seed protein and further purification methods,To improve the coagulation efficiencies of Moringa seed protein.Expand its scope of application.The best extraction method of Moringa seed protein in the present study can provide ideas and reference for the extraction and purification of Moringa seed active ingredients in water purification.
