Sedum aizoon is an endemic plant widespread in oriental countries, known as "Tu San-Qi", "Jiang xue san" in China. A crassulaceae genera perennial herb, has the function of hemostasis, removing blood stasis, resisting oxidation, resisting tumor, lowering blood pressure, reducing blood fat and so on. In this paper, we mainly study the ultrasonic assisted extraction of total flavonoids in Sedum aizoon L., and the optimization of response surface method of the extraction purification technology of macroporous resin and silica gel column, on the isolated samples of HPLC, UV, LC-MS research and analysis. Their antioxidant activities were also studied.The experiment temperature (℃), experiment ethanol concentration (%) and solvent to solid ratio were selected as the optimization response surface condition.The best experiment conditions were as follows:the experiment time was 20min, the experiment temperature was 57℃, and the experiment ethanol concentration was 80%, the solvent to solid ratio was 20:1. Through the validation experiment, the extraction content of flavonoids was 24.87±0.26mg/g.Through the static adsorption experiment, the best purification resin was selected as the D-101 macroporous adsorption resin,The effects of different pH values on the adsorption and resolution and gradient elution test were also investigated. The results showed that with the concentration of 30% and 50% ethanol, the elution effect was the best, can be eluted from the majority of flavonoids. Through repeated separation and collection of silica gel column to get the monomer coponent, through the HPLC, UV and LC-MS detection and reference to quercetin standard, it is speculated that the monomer material may be quercetin derivatives.The DPPH method was study on the antioxidant capacity of the extractions of the extracts of the initial solution of the ultrasonic wave, the resin and the purification the antioxidant capacity of the monomer coponent, ascorbic acid was used as a standard. Study found that:liquid weakest antioxidant capacity of the beginning of the ultrasonic extract,30% and 50% ethanol elution part of antioxidant capacity than ultrasonic primary extract is stronger than that of, the monomer portion of antioxidant ability is very strong, when the concentration reached 2mg/mL, the DPPH free radicals clear rate is equivalent to ascorbic acid, achieve 94%.
