| Lanolin resources are very rich in China, each year about 2-3 million tons but the utilization rate only about 10%, other were discharged directly except for a few applications in cosmetics industry or foreign acquisitions. It not only causes the great loss of resources, but also caused pollution to the environment. Therefore, it is necessary to strengthen the comprehensive utilization of lanolin and lanolin extracting from wool wastewater. Lanolin is cheap, only 1 million RMB per ton, which contains a large number of high value of cholesterol, so the separation and extraction of cholesterol from lanolin can solve problem of lanolin utilization rate of the smaller, and can complete the cholesterol of industrialization.In this paper, it is based on domestic and foreign research the extracting of cholesterol from lanolin, established the method that lanolin through saponification, the transformation of calcium soap,column chromatography and crystallization for extracting cholesterol, then conduct the pilot and cost accounting and the results as follows:l.To optimize the conditions for the saponification reaction and transformation of calcium soap, and ultimately determine the 28.35 g lanolin the optimum conditions of saponification reaction for 3gNaOH, 150mL ethanol and 80℃ for reaction 6h. The optimal conditions for the separation of the sodium soap and the sodium chloride on the transformation of calcium soap are 4.17 g calcium chloride,35℃. Under the optimum conditions, the yield of cholesterol could reach 96%, and the purity was 22.1%.2.The process route of extracting cholesterol from wool alcohol by column chromatography was determined. The silica gel was static phase, petroleum ether and ethyl acetate as mobile phase. A set of optimum conditions of column chromatography were optimized:the sample amount of 15 g (load 55 mg/g), column height of 25 cm, column diameter of 5cm, velocity of 15mL/min, silica gel mesh of 300-400, cholesterol were qualitative analysis (IR, NMR) and quantitative analysis (gas chromatography, liquid chromatography), column chromatography step in cholesterol production rate 92.4%,94.2% purity was measured.combined with saponification step, the total yield of extraction of cholesterol in lanolin was 88.7%, the purity of 94.2%.3.Successful to extract cholesterol from the cholesterol in the pilot amplification, the process of the step enlarged for 53 times, column chromatography step amplification 8 times, the extraction of cholesterol yield of 83%, the purity of 95%. Then the cost accounting with annual processing 1000 tons of lanolin calculated; 10.5 million cost of raw materials, electricity of 50000, equipment depreciation costs of 56 million, factory and install costs of 40 million,375 million workers wages, energy consumption of 375 million, sales income of 25.28 million, deduction of enterprise income tax of 25%, the profits due to 453.75 million.4.The reason for the large amount of solvent consumption is that the solubility of the solvent is lower, the column height is increased, change the ratio of petroleum ether and ethyl acetate in the solvent can greatly reduce the consumption of solvents. |
