Surface Plasmon Resonance (SPR) sensing technology has been widely applied in the areas of life science, environment monitoring and food safety for its merit of no label, high sensitivity, portability and real-time and real-scene ability. In recent years, the development of SPR sensing and instruments keeps stable. The detection accuracy of the traditional angular-interrogation SPR sensor cannot gradually satisfy the needs of analysis and detection, since there are some bottleneck and limitation when it detects some analytes with low concentrations or some micromolecules. Therefore, the thesis is aimed at enhancing the performance of the SPR sensor. Based on our home-built multi-channel SPR experiment instrument, novel metal film systems are proposed to improve the performance of our SPR sensor. Main contents and achievements of this thesis include:1. A microwell gold configuration has been studied in order to improve performance of the SPR sensor. The microwell gold configuration is simulated using FDTD Solutions, which can provide some therotical basis. The configuration is fabricated and its performance is evaluated through a refractometry experiment with sodium chloride solutions. According to the experimental results, the refractive index resolution is 4.7×10-7 RIU. Compared with the planar gold configuration, its performance is improved by a factor of two.2. A bimetallic configuration with gold and silver that can improve the performance of the SPR sensor has beem studied. The configuration is simulated using MathCAD, LabVIEW and FDTD Solutions and the optimium parameters are obtained. The bimetallic chip is fabricated and its performance is evaluated by a refractometry experiment. The refractive index resolution is 5.3×10-7 RIU, which shows that this sort of configuration can improve the performance of the SPR sensor by a factor of two compared with the planar gold configuration.3. The cadmium ion, a kind of heavy metal ion, is detected using multi-channel SPR experiment instrument and the bimetallic configuration. Based on the interaction between the bovine serum albumin (BSA) and cadmium ions, satisfactory results are obtained by a simple experiment without regeneration and the detection limit is 0.01?M.
