Phenolic acids not only has strong ability of clearing free radicals and antioxidant activity but also has the function of anticancer and anti-inflammatory. In order to develop and utilize its potential value, it is necessary to search the separation and purification method of phenolic acids. Magnetic nanoparticle materials have show great application potential in the field of separation because of their simple preparation process, good biocompatibility, unique magnetic response property, advantages of environmental protection and recycling, etc. As a support-free liquid-liquid chromatographic technique where the mobile and stationary phase are composed of two immiscible solvents or solutions, high speed countercurrent chromatography has so many advantages, such as economical, high capacity with an excellent product purity. It has been a vigorous development method of separation and purification of active substance from natural products.In this paper, based on online high speed countercurrent chromatography, two novel process were established, aiming at the separation of phenolic acids. Based on high speed counter current chromatography, the aim of this paper is to design and synthesize the functionalized magnetic nanoparticles with strong adsorption of phenolic acids. Two kinds of functionalized magnetic nanoparticles which have strong adsorption effect Fe3O4@SiO2@Cn and Fe3O4@SiO2@NH2 were designed and synthetized. Adsorption rate reached more than 98%. A novel reliable on-line solid phase extaction and high speed counter current chromatography process for purification of three kinds of phenolic acids from sugarcane skin has been developed. The on-line separation conditions including the saturated extent, elution mode were optimized. Finally, three phenolic acids including 6.73 mg of gallic acid,10.85 mg of p-coumaric acid and 2.78 mg of ferulic acid were obtained with purities of 60.2%,95.4%, and 84.0% respectively by using this online process from this 150 mg extracts.
