Gomphidius Rutilus as a rare edible fungus, has not been effective developed and utilized. The current research of Gomphidius Rutilus mainly focuses on the polysaccharides, but other biological activities has been fewer reported. In this paper, the optimum conditions of total flavonoids extraction form Gomphidius Rutilus were investigated, and the purification of total flavonoids of Gomphidius Rutilus(GRF) using macroporous resin was discussed. The compositions of GRF were confirmed by high performance liquid chromatography(HPLC) and infrared spectrum. The antioxidant capacity of GRF in vitro was also studied. This study provides references for further development and utilization of Gomphidius Rutilus.The total flavonoids were extracted from Gomphidius Rutilus using ethanol reflux method. Extraction time, liquid ratio of volumetric fraction to ethanol, and extraction temperature were selected as single factors. Also the optimum conditions of extraction were investigated through response surface methodology. The results were as follows: 83%ethanol volume fraction, ratio of water to raw material as 15; extracting time as 2.5 h;extraction temperature as 85?. The total flavonoids extraction yield was 6.72% under this condition.The absorption of total flavonoids was discussed by four kinds of macroporous adsorption resin(AB-8, D101, silica gel and polyamide). Through the static adsorption and desorption experiment, this topic compared the four different types of macroporous resin adsorption performance of total flavonoids. Results showed that the polyamide resin has better comprehensive performance and higher rates of adsorption and desorption. The best condition of dynamic adsorption were as follows: the best concentration of the sample as1.0-1.5 mg/mL, solution pH as 5, on the sample flow rate as 0.6~0.8 BV·h-1, elution solvent as 60%, elution velocity as 1.0 BV·h-1. After stress concentration and freeze drying, the purity of total flavonoids can reach 70.18%.Total flavonoids of Gomphidius Rutilus inclued baicalin through UV wavelength scanning, infrared spectrum, and high performance liquid chromatography(HPLC).The antioxidant capacity of GRF in vitro were studied by the reducing force, scavenging1, 1-diphenyl bitter phenylhydrazine free radical(DPPH·), the ability to remove hydroxyl free radical(·OH), and removing superoxide anion radical(O2-·) ability. Results showed that the oxidation resistance and concentration appear good concentration-response relationship, with the increase of concentration, oxidation resistance increase gradually.
