Fucoidan is a bioactive polysaccharide, so it is widely studied recently. The Kjellmaniella crassifolia from Japan was taken as raw material,Crude fucoidan F was extracted and then five fractions were obtained by anion-exchange chromatography on DEAE-Sepharose Fast Flow. The contents of sulfate and polysaccharide in five groups were determined respectively, the monosarcharride composition of five fractions by HPLC and the antitumor activities were analyzed as well. The results and conclusions are as follows: 1.The SO42- contents of F-0, F-1, F-2, F-3 and F-4 determined by Ba Cl2 method were 15.84%, 3.95 %,16.87 %, 21.69 %? 26.11 % respectively. Their total polysarccharide contents of F-0, F-1, F-2,F-3 and F-4 were 45.52 %,32.43 %,53.26 %,46.88 %,64.25 %, respectively. 2.The findings of the monosaccharide composition of these fractions by HPLC showed that, the monomers were fucose, galactose, mannose, xylose mainly. However, the composition ratio of monosaccharides in each fraction was different. The mannose was the main monomer of F-1, which was28.84%, and the fucose was the main compositon of F-4, which was up to 91.44%. 3.The results for inhibiting human Hep G2 cell, abdominal macrophage RAW264.7, human colon cancer HT-29 cell and lung cancer A549 cell growth in vitro indicated that F-2 showed a strong inhibitory effect on liver cancer of Hep G2 cell, F-3 illustrated quite strong inhibiting effect on abdominal macrophage cell and human colon cancer HT-29 cell, and F-0 showed strong inhibiting effect on lung cancer A549 cell.
