This paper tries to hydrolyze fucoidan by HCl,and showing its effects of antitumor and immunomodulatory.1.Fucoidan F2 was degraded by 0.1 mol/ml hydrochloric acid,and abtained F2-a,F2-b and F2-c.Chemical composition analysis indicated that F2-a?F2-b?F2-c contained 88%?26.2%?54% total polysaccharides,with 6.55%?56.68%?1.87% SO42-;HPLC analysis indicated that F2-a were mainly composed of mannose and glucuronic acid,F2-b were composed of mannose?glucuronic acid and fucose,F2-c was just fucose;The results of IR spectrum indicated that absorption peaks of O-H(3369-3403cm-1)in the F2-a and F2-b,;S=O(1144cm-1)existed F2-b,which demonstrated that F2-b contain sulfate substitute.13 C NMR spectrum indicated thatthe displacement of C2-C5 was 62-75 ppm of F2-a?F2-b and F2-c,and 18.13-18.73 ppm is the displacement of C6 of F2-b and F2-c.2.The result for inhibiting HepG2 ? MCF-7 ? Hela ? A549 ? HT29 and RAW264.7 growth in vitro indicated that inhibition ratio of F2-b on HepG2 was showed 36%,F2-c showed with inhibition ratio 32% on Hela,the inhibition ratio of F-2 and F2-c on RAW264.7were 43.26%?42.12%,respectively at 800 ?g/ml;no effect on MCF7 in all samples.3.The results of MTT and Elisa Assay showed that effect of F2?F2-a?F2-b and F2-c on RAW264.7.The contents of secretion of cytokines TNF-? ? GM-CSF of F2 ? F2-a were 27.131 ng / L and 286.96 ng / L,respectively at the concentration of 20?g / mL.
