Curculigo is the roots of Curculigo orchioides Gaertn. In this paper, some polysaccharide fractions were isolated, purified from curculigo crude polysaccharides. The physicochemical and structural characteristics of the polysaccharides were studied by using chemical and instrumental analytical methods, The main contents are as follows:1. The curculigo crude polysaccharides were isolated and purified by deprotein. DEAE-Cellulose and Sepharose CL-6B column chromatography, four polysaccharide fractions were obtained, named as XMP-1~XMP-4. Their homogeneity were determined by gel colum chromatography and gelpermeation chromatography(GPC), the results showed that XMP-1 was homogeneous polysaccharides, while XMP-2 was, inhomogeneous. So XMP-1 was taken studied its physicochemical and structural characteristics.2. The monosaccharide compositions was investigated by GC-MS. The molecular weight(Mw)was identified by GPC. The uronic acid was identified method of m-hydroxydiphenyl and carbodiimide reduction.XMP-1 was composed of mannose, glucose, galactose with a molar ratio of 81.2:5.7:1. The total carbohydrate content of XMP-1 was 91.8% and the molecular weight was 3031 Da. XMP-1 is a neutral pure heterosaccharides.The IR spectrum showed that XMP-1 have the typical characteristic absorption peak of polysaccharides, and XMP-1 contain ?-D- mannopyranose. The UV spectrum showed that XMP-1 has no nucleinic acid, protein and pigment.3. The stucture of XMP-1 was analyzed by Partial acid hydrolysis, Periodate oxidation, Smith degradation, Methylation and Congo.XMP-1 has a backbone and mainly consisting of mannose, glucose, galactose. the backbone margin and the branch of XMP-1 mainly consisting of mannose, glucose. The terminal-residues of backbone mainly consisting of mannose, glucose, galactose. XMP-1 has the following structure characteristics. It has a backbone and mainly consisting of 1,4- Glc 1,3-Man 1,6-Gal 1,6-Man1,2,6-Man and 1,3,6-Man. The bachbone terminal-residues of XMP-1 contains 1-Glc, XMP-1 has a few branch contain 1,3-Man, 1,6-Man, 1,6-Gal, 1,4-Glc, with 1,2,6-Man and 1,3,6-Man as branched chain connection.The Congo red test showed that XMP-1 didn't exist the triple-helical conformation in water.
