As an important component of nutrients,polysaccharides are widely existing in microorganisms,plants and animals,Plant polysaccharides have been studied for a long time due to their low toxicity,high biocompatibility,high biodegradability and broad-spectrum therapeutic effect.The research of Loranthaceae is more focused on the flavonoids,and agglutinin,the study of polysaccharides,extract of Loranthaceae,needs to be deeper discovered besides the study of Surrula parasitica L and the polysaccharides of Loranthus delavayi Van Tiegh,which can be parasitized on Camellia oleifera plants.The purpose of this study was to extract,purify and identify the polysaccharides of M to obtain the homogeneous components of M,to analyze the structure of polysaccharides of M and its homogeneous components and evaluate the biological activities?antioxidant and ancoagulant abilities?,it is expected to provide the experimental basis for the development and utilization of M.Six kinds of crude polysaccharides were extracted from the branches and leaves of M with hot water,hydrochloric acid and sodium hydroxide as solvents and then the purified polysaccharides were separated and purified by DEAE-SephadexA-25 ion exchange column.The purified polysaccharides were identified by Sephadex G-100 gel column chromatography and Ultraviolet spectroscopy,and eleven kinds of one-component polysaccharides were obtained.The contents of total sugars,uronic acids and proteins of the seventeen polysaccharides were determined by phenol-sulfuric acid method meta-hydroxybiphenyl method and Coomassie brilliant blue method.The monosaccharide composition and average molecular weight of seventeen kinds of polysaccharides were measured by gas chromatography?GC?and gel permeation chromatography?GPC?.The main structural characteristics of seventeen kinds of polysaccharides were measured by infrared spectroscopy?IR?,ultraviolet spectroscopy?UV?and Congo red test.The antioxidant capacity of seventeen kinds of polysaccharides in vitro was tested by ABTS and DPPH free radical scavenging experiments.The anticoagulant activity of seventeen kinds of polysaccharides in vitro was tested by APTT?TT and PT,and the relationship between extraction site,extraction solvent,polysaccharides structure and antioxidant and anticoagulant biologiacal acticoagulant biological activity in vitro was discussed.The results show that:?1?the total sugar content?phenol-sulfuric acid method?,uronic acid content?meta-hydroxybiphenyl method?and protein content?Coomassie brilliant blue method?of the seventeen polysaccharides were not identical.?2?Most of the seventeen polysaccharides are composed of ribose,rhamnose,arabinose,mannose,glucose and galactose,and their monosaccharide contents and molar ratios are different;?3?The molecular weights of seventeen polysaccharides are different and the polysaccharides extracted with sodium hydroxide solution as sovent had smaller molecular weight;?4?Infrared spectrum analysis showed that the eleven kinds of polysaccharides were?configuration with pyranoseand and six kinds of were?configuration with pyranoseand.?5?Bioactive experiment results showed that seventeen kinds of polysaccharides have scavenging free radicals and anticoagulant in vitro,with sodium hydroxide solution as solvent extraction polysaccharides and its purification uniform components cleared ABTS radical highest titer the EC50 of about20 mg/L,seventeen kinds of polysaccharide mainly endogenous clotting pathways play an anticoagulant activity.The anticoagulant and antioxidant activities of polysaccharide of M extracted with sodium hydroxide solution are better,which may be related to the factors suchn as low molecular weight,?configuration with pyranoseand and high galactose and glucose content.This study is expected to provide experimental basis for the development of polysaccharides of Mistletoe of camellia oleifera.By studying the polysaccharides of M,the medicinal value of polysaccharides of M can be predicted,and this study is expected to provide experimental basis for the development of polysaccharides of M.
