Seabuckthorn(Hippophae rhamnoides Linn.),is a kind of deciduous shrub belong to Elaeagnaceae Hippophae.Seabuckthorn are mainly distributed in the arid region in China.The production of Shanxi province is the largest.Seabuckthorn output a large number of seabuckthorn seed residues in the course of processing.The protein content of the seed residues is 25.0%-30.0%,however this part of the protein is not fully reasonable used.In this study,the residue that left after supercritical carbon dioxide extraction of seed oil from sea buckthorn seeds was the raw material,and the protein is changed into a polypeptides with anti-alcohol and bacteriostasis effect by enzymatic hydrolysis,which was intend to provide new ways for the development of the comprehensive utilization from seabuckthorn seed.The research contents of this paper are as follows:1.Preparation of seabuckthorn seed anti-alcohol polypeptides.Used seabuckthorn seed as raw material,polypeptide yield and ADH activity as the index to determine papain protease as the optimum enzyme to prepare active polypeptides.Through combination of single factor and orthogonal array designs an enzyme dosage of 4000 U/g,an initial pH of 6.5 and 3.5 h hydrolysis at 50? were found to be optimal to obtain the maximum ADH activity.And the ADH activity and peptide yield of polypeptides was 17.56%,38.80%under the above conditions.Of those,the effect of pH on ADH activation rate was significant,next is the time,last is the temperature of enzymatic hydrolysis.2.Research on isolate and purify active component from seabuckthorn seed polypeptides.The molecular weight in 500-2000 Da peptide had the highest ADH activation rate by membrane separation,and the rate was 57.52%.Next,the peptide with molecular mass between 2000-3000 Da also had ADH activation rate,which was 35.09%.The purity of peptide from Seabuckthorn seed is improved by Glucose-lowering.The reducing sugar content reduced from 0.77%to 0.29%,and the ADH activation rate is increased by 7.12%.3.Study on the auxiliary anti-alcohol function of seabuckthorn seed residue active polypeptides.By animal experiments,auxiliary anti-alcohol function of seabuckthorn seed active polypeptides on the mice were researched.The results show that polypeptides 100 and 400mg/kg · bw dose group of blood ethanol levels were significantly lower than those in the model control group.And sober time,drunk time also reduced,so the active polypeptides had a good auxiliary anti-alcohol effect,and the greater the dose,the better the results,there was a certain dose-response relationship.4.Antibacterial activity of enzymatic hydrolysates of seabuckthorn seed residue.Using protamex and pepsin for enzymolysis,and using the method of membrane separation for purification.Then get the enzymolysis polypeptide with antibacterial activity.Results indicated that,protamex hydrolysate possesses bacteriostasis in vitro of different degrees on Staphylococcus aureus,Salmonella,Pseudomonas aeruginosa.And pepsin hydrolysate possesses bacteriostasis in vitro of different degrees on Escherichia coli,Staphylococcus aureus,Salmonella.Result of animal oxidation experiment showed that,Salmonella bacteria in mice feces were extremely significant(p<0.05)compared with the model group,enzymolysis polypeptide was given by gavage.Staphylococcus aureus bacteria in mice feces were extremely significant(p<0.05)compared with the model group,enzymolysis polypeptide was given by gavage.
