Seabuckthorn resources are rich in our country,seabuckthorn fruit include a variety of vitamins,trace elements and biological active matter,and it's a medicine edible plants.seabuckthorn processing in China is at the emerging stage,and mainly to shallow level processing of seabuckthorn fruit,seabuckthorn seed residue waste caused by abandoned.This thesis use seabuckthorn juice ? oil ? seed as raw material,comprehensive utilization of seabuckthorn.First use of seabuckthorn seed preparation into seabuckthorn protein,seabuckthorn polypeptide are obtained by enzymatic digestion seabuckthorn protein,then add seabuckthorn polypeptide and seabuckthorn oil into seabuckthorn juice,study the emulsion stability,Finally add taste masking agent for sensory evaluation.finally making into rich nutrition,sweet and delicious seabuckthorn polypeptide oral liquid.The main research contents and results of this experiment are as follows :(1)Determine the optimum process of seabuckthorn polypeptide preparation.First production of seabuckthorn peptide standard curve for : Y = 0.176 + 0.0007 x,correlation coefficient R2 = 0.9981.The substrate is extracted by our laboratory seabuckthorn protein,from papaya protease,alkaline protease,neutral protease and bromelain to determine the best protease,the result is alkaline protease and neutral protease.The two compound protease,and determine the proportion of compound protease for alkaline protease and neutral protease 2:1,on the basis of single factor experiment.After enzymolysis seabuckthorn polypeptide content as evaluation index,with enzyme solution p H,digestion temperature,digestion time and compound enzyme additives for indexes orthogonal test,determine the optimum extraction process for each target : enzyme solution temperature of 55 ?,compound enzyme amount of5000 u/m L,enzymolysis time for 4 hours,enzyme solution p H 7.0.(2)Seabuckthorn oral liquid emulsification and stability study.According to the principle of similar miscibility choose two standard emulsification(Tween-80 and 2,3-Dihydroxypropyl octadecanoate).According to the HLB value scope of 6 ? 14 design series formula,to mix various HLB values of emulsifier,emulsifying ability is measured by turbidity method respectively to determine the best emulsifier types and composite emulsifier formula is:Tween-80 and 2,3-Dihydroxypropyl octadecanoate 1:1.Pectin,carrageenan and xanthan gum,sodium carboxymethyl cellulose(CMC)and guar gum several stabilizers to join this system,determine the best stabilizer is xanthan gum.On the basis of single factor experiment,to stabilize the value(ES)as evaluation index,the seabuckthorn oil amount,stabilizer concentration and compound emulsifier concentration effect on the stability of the seabuckthorn polypeptide oral liquid.Three factors three levels of response surface experiment,the effects of three factors,determine the major and minor in C > A > B,namely the seabuckthorn oil amount > stabilizer composite>emulsifier content.Optimizing the best recipe for : stabilizing agent content is 0.3%,the composite emulsifier content is 0.4%,seabuckthorn oil content is 1%.(3)Seabuckthorn nutrition liquid of sensory evaluation and the choose of the sterilization.On the basis of single factor,the contents of honey,xylitol contents,seabuckthorn peptide contents as test factors,Designed nine groups of the orthogonal experiment.In an integrated sensory score as an index of sensory evaluation,the most suitable adding amount of each material is : honey adding amount was 0.5%,xylitol adding amount was 0.3%,the content of seabuckthorn polypeptide extract was 15%.Will deal with good seabuckthorn peptide oral liquid with the method of pasteurization,slightly change the sterilization temperature and sterilization time.Pasteurization temperature design three groups according to the actual conditions,the determine of the total number of colonies,eabuckthorn peptide contents,sensory evaluation score and seabuckthorn peptide oral solution before and after sterilization,ultimately determine the best sterilization conditions for 85 degrees Celsius,the processing time for 5 minutes.
