Collagen is one of the most widely distributed protein in organism,it has good biological properties,such as safety,low immune activity,biocompatibility,application in food,medical materials,cosmetics and other fields widely.Even so,the natural collagen material still has many defects,such as tensile properties,thermal stability is not ideal.Collagen fiber after the restructuring of glue is one of the main form of collagen material application,the application of collagen gel properties and material range and application effect are closely related.This paper focus on research on collagen gel properties and its influencing factors,including: gelatination formation method,drying method and the collagen fiber reorganization of the gel and other factors.By adopting different methods for preparation of collagen gel after conducting gel performance analysis,explore the influencing factors on the rheological,quality and structure of the gel properties and biological properties of impact,for the development and application of new collagen gelation materials provide new theoretical support.In pigskin enzyme soluble collagen(pigskin PSC)as raw material,using different sample preparation method for the preparation of collagen gel(fiber restructuring rubber directly swelling,extraction of collagen sponge rubber and fiber restructuring swelling rubber sponge),and measure its including swelling properties,thermal stability,performance of cell biology,physical and chemical properties,using the method of SEM characterization of gel microstructure,to probe the differences of different collagen gel on the performance and structure.The experimental results show that the swelling rate of collagen gel was followed by the size of the direct swelling gel(253.14 ±21.08%)> Fiber restructuring gel(202.34 ± 20.41%)> Fiber-freeze-dried swelling gel(185.27 ± 23.89%);The thermal stability properties of the gel(denaturation temperature,Td)followed by Fiber-freeze-dried swelling gel(45.78 ?)> Fiber restructuring gel(44.13 ?)> direct swelling gel(42.56 ?).Promote the cell proliferation ability are: Fiber restructuring gel > Fiber-freeze-dried swelling gel > direct swelling gel.Gel microstructure SEM analysis results show that the three kinds of average pore diameter size of collagen gel Fiber-freeze-dried swelling gel(343.41 ± 56.32 um)> Fiber restructuring gel(294.37±23.22 um)> direct swelling gel(123.53 ± 44.72 um),restructuring and fiber plastic than the other two kind of glue are uniformly pore size and arrangement more closely also.And HE staining to observe the internal structure of collagen material also found that fiber reorganization compared with other two kinds of adhesive space structure isrelatively uniform.Adopts the structure of the gel fiber fixed SEM observations show that can only be observed net-like fibre restructuring plastic fiber configuration,fiber diameter around 105.03 nm,while the other two kinds of adhesive are closely packed into pieces.The experimental results show that different collagen gelation method directly affect the collagen gel properties and gel microstructure.From pigskin PSC as raw material,using different methods of drying fiber restructuring swelling rubber sponge(liquid nitrogen to freeze drying glue,conventional freeze drying,vacuum drying adhesive,drum wind drying glue),respectively,and the determination of the various physical and chemical properties and performance of cell biology and gel microstructure,studying the effect of drying methods on the properties of gel structure.The experimental results show that: four drying methods products gel swelling rate of liquid nitrogen to freeze drying(198.33± 5.02%)> conventional freeze drying(185.21±17.94%)> blast drying(149.93±19.80%)> vacuum drying(123.12 ±23.42%);Thermal stability(the denaturation temperature,Td)followed by vacuum drying(52.34 ?)> drum wind drying(49.53 ?)> conventional freeze drying(46.65 ?)> liquid nitrogen to freeze drying(43.28 ?).To promote cell proliferation ability are: liquid nitrogen to freeze drying >conventional freeze drying >drum wind drying>vacuum drying.The gel microstructure SEM analysis results show that the order of four kinds of gel and average pore diameter size is conventional freeze drying(316.45 ± 18.51 um)> liquid nitrogen to freeze drying(283.65 ±37.28 um)> drum wind drying(286.32 ± 25.42um)> vacuum drying(149.15 ±8.65 um),compared with conventional freeze drying and vacuum drying the other two kinds of gel pore size is uniform.And HE staining to observe the internal structure of collagen material also found that compared to conventional freeze drying of other three kinds of glue space structure is relatively uniform.Adopts the structure of the gel fiber fixed SEM observations show that four of the drying method,the collagen material after swelling all could not see the original fiber structure,which are closely packed into pieces.The experimental results show that different ways of drying performance will directly affect the collagen gel and gel microstructure.In grass carp PSC and pigskin PSC as raw materials,the use of dynamic rheometer as the main research means,in different source collagen blend fiber restructuring dynamics monitoring and gelation product performance analysis.Fiber restructuring dynamics research found that,in ensuring that collagen assemble into fiber temperature range,pigskin and grass carp PSC assembly process are influencedby temperature,temperature,fiber and then faster restructuring.At the same time,the two types of collagen product fiber pyrolysis temperature also has difference,grass carp PSC assembly fiber dissociation temperature is 41.52 ?,significantly lower than the pigskin PSC fiber dissociation temperature of 45.05 ?.Two types of collagen fiber after blending restructuring dynamics analysis results show that compared with their own corresponding single collagen fiber restructuring process,blending collagen fibers after restructuring dynamics behavior has changed significantly,blending fiber reorganization of the elastic modulus value and viscosity of the gel modulus value in collaborative,prompt a blend between the two types of collagen fiber restructuring behavior.Further carry out the restructuring of the fiber structure and gel analysis showed that the blend fiber restructuring and single collagen fiber in the fiber diameter,D cycle characteristic value,the respect such as gel pores size difference is obvious.At the same time,the blend fiber restructuring gel product also show good capability of promoting cell proliferation.The experimental results show that the two kinds of different sources of collagen can occur together fiber restructuring behavior,and the fiber reorganization of the product structure and gel properties change.The study results suggest,by different source blend between collagen fiber reorganization,can rise to adjust and change the effect of collagen gel properties.
