In this paper, we take the corn starch as raw material, pulp to mass percent concentration of 50%, through single factor and response surface experiments optimization of the best alpha- medium temperature amylase, high temperature resistant alpha amylase step by step liquefaction process, through single factor experiment and two times orthogonal rotation combination design the optimum amount of enzyme saccharification best amyloglucosidase, beta amylase and pullulanase combination. The research results as follow:(1) With the thermostable alpha amylase temperature alpha amylase and anhydrous calcium chloride as the research, the adding amount, the liquefaction temperature and the time of liquefaction were variable, set contrast test, with DE value as a measure index, optimal liquefaction conditions of high concentration corn starch milk were determined. The main operating process as follow: Mass percent concentration of 50% high concentration corn starch milk at 80 centigrade, add 14U/g medium temperature alpha amylase, mass percent concentration of 0.15% anhydrous calcium chloride solution, liquefaction 20 min, and then the temperature increased to 93, adding 11U/g thermostable alpha starch, liquefaction 20 min, corn starch milk processed by the steps can meet the needs of the next step in the saccharification reaction.(2) The main research on amyloglucosidase, beta amylase and pullulanase enzymes for three, through single factor experiment and orthogonal rotation combination test, get the optimum parameters of three kinds of glucoamylase saccharification synergy, the establishment of the corn starch saccharification reaction system with high concentration of liquefaction products. Optimum parameters for saccharification: The amount of enzyme beta amylase was 21.82U/ g, pullulanase enzyme 0.354 U/g, amyloglucosidase enzyme 59.55U/g, saccharification temperature is 60, saccharification time for 40 hours.
