DNA is the carrier of genetic information in all organisms.DNA is the main target molecule for most of small molecules which can interact with and damage template function of DNA,break the chain of nucleic acids and therefore affect the gene regulation and expression.Interactions of small molecules with DNA are investigated to understand bind of DNA with protein,anticancer mechanism and to design and synthesize new drug.Due to characteristics such as better binding porperties of DNA,phenazine derivatives can provide new design ideas for the development of novel anti-tumor and anti-cancer drugs.In recent years,the engineering in medicine and biology fields,we found that interaction with DNA as a molecular probe,there are fluorescence markers to produce good results,they has enormous potential application value in this regard.Methylene violet 3RAX and its derivatives is a kind of phenazine compounds that take electric charges.They can be widely used in medicine,material science,dye industrial,and so on.At the same time,their water solubility can effectively impove the activity of antitumor drugs.This paper is going to introduce the synthesis of the methylene violet 3RAX and its derivatives,they are involved to demonstrate their interaction with DNA.All these are treated as the basis of further research on the exploration of new kinds of phenazine antitumor drugs.This thesis includes mainly:1.The development in the studies of DNA,including sturcture of DNA,interaction of DNA with other matter and the mothods about interaction of DNA and small molecule was described.The evolution of phenazine compounds,methylene violet 3RAX,indomethacin is introduced briefly,especially in the application of biology and medicine,and so on.At the same time,the anti-tumor mechanism of phenazine compounds was also described.The design philosophies of this thesis are brought forward on these bases.2.Methylene violet 3RAX derivatives were synthesized by retaining the mother ring structure,transforming the surrounding groups,being tailed with bioactive molecules,such as indomethacin,and their structures were confirmed by 1H NMR,IR,MS.3.The binding properties of methylene violet 3RAX derivatives to calf thymus(ct-DNA)were investigated by using absorption,fluorescence and circular dichroism(CD)spectra.The ability ofmethylene violet 3RAX derivatives to generate singlet oxygen were investigated by using DPBF.The antitumor activity of these derivatives are predicted on the base of these test data.4.The conclusion of this thesis,the work still undergoing and the purpose and expectation of our future work.
