Prodigiosin is secondary metabolites of methoxypyrrolo skeleton structure which is consisting of three pyrrole ring.It is widely concerned due to its good anticancer vitality.At the moment,,prodigiosin is mainly detected by spectrophotometry.The experiment adopts the mobile phase solution of methyl alcohol and water to achieve fast and accurate determination of prodigiosin in fermentation broth,making use of liquid chromatography of high performance(HPLC)to detect prodigiosin content,meanwhile obtaining a small amount of sample with high pureness by using liquid chromatography of preparative high performance(PHPLC).The intent of this mission is to study the dynamic process of silica gel column chromatography purifying prodigiosin so as to give a guideline of using PHPLC to purify prodigiosin with higher purity.(1)By investigating adsorption performance of prodigiosin,it was found that adsorption isotherms for prodigiosin is fit with Langmiur model,the correlation coefficient R2 is 0.9866,elution profiles can be better fitting prodigiosin elution process,the correlation coefficient can reach 93.45%.(2)Based upon studies of prodigiosin kinetics,the best conditions for PHPLC was investigated by exploring flow rate,sample volume,temperature and other conditions on HPLC performance.Optimum conditions for SB-C18(9.5mm×2.5dm,5×10-3mm)PHPLC are solution(methanol:water =51:49),2×10-3 L/min of rate,2×10-5 L of the sample volume,25? of temperature.Prodigiosin preparation purity by PHPLC used is high.
