Research purpose:Research on the effects of different ratios of DHA/EPA in intervening lipid metabolism of high fat mice and explore the possibility and mechanism of the intervention.Research contents:90 male ApoE-/-mice,(6 weeks old,with a weight of 18±3g),who are divided into 6 groups randomly: ND group is fed with common feedstuff,and intervened by gavaging with distilled water;HFD group is fed with high fat feedstuff,and intervened by gavaging with distilled water;ALA group is fed with high fat feedstuff and intervened by gavaging with ALA at 1mg/g bw;DHA/EPA=2:1 group is fed with high fat feedstuff,and intervened by gavaging at 1mg/g bw when DHA/EPA=2:1;DHA/EPA=1:1 group is fed by high fat feedstuff,and intervened by gavaging at 1mg/g bw when DHA/EPA=1:1;DHA/EPA=1:2 group is fed with high fat feedstuff,and intervened by gavaging at 1mg/g bw when DHA/EPA=1:2.Kill the mice after feeding them for 12 weeks,and measure the lipid level inside the body of each group of mice: TC concentration and TG concentration in serum,TC concentration and TG concentration in liver,composition of fatty acid in serum and composition of fatty acid in liver.Research results:(1)High fat diet may cause abnormal lipid metabolism of male ApoE-/-mice,make the TG concentration and TC concentration in their liver and serum higher than normal level;make the composition of fatty acid in liver and serum abnormal.All of the ALA and different ratios of DHA/EPA intervention groups can adjust and control the lipid metabolism of the male ApoE-/-mice,the effect of PUFAs on the regulation of TG,TC content and fatty acid composition in serum and liver of high fat mice was the best in DHA/EPA=1:2 group.(2)All of the ALA and different ratios of DHA/EPA intervention groups can adjust and control the enzymatic activity of relevant enzyme of lipid metabolism of the male ApoE-/-mice: inhibit the enzymatic activity of relevant enzyme of lipid metabolism synthesis such as ACC-1 and FAS in liver of male ApoE-/-mice and enhance the enzymatic activity of the CPT-1 enzyme used for decomposing key genes in lipid metabolism.The regulation effect of PUFAs on the enzymatic activity of the key enzymes of lipid metabolism in the liver of high fat mice was the best in DHA/EPA=1:1 group.(3)All of the ALA and different ratios of DHA/EPA intervention groups can adjust and control mRNA the expression of relevant genes of lipid metabolism of key genes of lipid metabolism of the male ApoE-/-mice: inhibit the mRNA expression of relevant genes of lipid metabolism synthesis such as SREBP-1C,ACC-1,FAS and SCD-1,and promote the mRNA expression of relevant genes of lipid metabolism decomposition such as AMPK,PPAR-?,CPT-1 and DGAT.The regulation effects of PUFAs on lipid metabolism related gene mRNA in high fat mice were the best in DHA/EPA=1:2 group.(4)All of the ALA and different ratios of DHA/EPA intervention groups can adjust and control the protein expression of key genes of lipid metabolism of the male ApoE-/-mice: inhibit the protein expression of SREBP-1C,the key gene of lipid metabolism synthesis,and promote the protein expression of key genes of lipid metabolism decomposition such as AMPK and PPAR-?.The regulation effects of PUFAs on protein expression in high fat mice were the best in DHA/EPA=1:2 group.
