During the development of diet-induced obesity,excessive energy intake will lead to adipocyte hypertrophy and hyperplasia,which results in insulin resistance,type 2 diabetes and hypertension.It has been reported that excessive adipogenesis and chronic low-grade inflammation in adipose tissues play a primary role in the development of obesity and obesity-related syndromes.Flavonoids have high chemopreventive efficiency,such as anti-inflammation,anti-cancer,anti-mutation.As an important dietary flavone,apigenin widely present in fruits and vegetables.However,its effects on diet-induced obesity and associated insulin resistance still need more systematic and further research.In this study,male C57BL/6 mice were divided into three groups and fed with low-fat diet(LFD),high-fat diet(HFD)with or without 0.04% apigenin(0.04 HAP)for 12 weeks,respectively.The results showed that apigenin reduced HFD-induced body weight gain,fat deposition and adipocyte hypertrophy.Meanwhile,apigenin improved obesity-related insulin resistance.Then we did further experiments to study the mechanisms of diet-induced obesity and obesity related insulin resistance both in vivo and in vitro.For lipid metabolism,apigenin not only inhibited adipogenesis genes PPAR? and SCD1 expression,but also activated SIRT1/FOXO1/ATGL pathway and increased lipolysis genes ATGL and HSL expression.Furthermore,apigenin markedly increased UCP-1 and PGC-1? genes expression which participate in thermogenesis.For inflammation,flow cytometry and Real-time PCR analysis showed that apigenin reduced adipose tissues macrophages accumulation and macrophages inflammatory polarization.And,this maybe achieve by inhibiting the nuclear factor-?B(NF-?B)and mitogen-activated protein kinase(MAPK)inflammatory signaling pathways.In conclusion,our study indicates that apigenin ameliorates diet induced obesity and obesity related insulin resistance partly through regulating lipid metabolism and inflammation polarization.Therefore,as a natural flavonoid,apigenin provides a new dietary strategy to obesity metabolic syndromes.
