In the present study,the BLF/TP/?-PL nanoparticles loaded with bamboo leaf flavonoids(BLF)were prepared by nanocapsule technology,using turnip polysaccharides(TP)as a carrier,?-polylysine(?-PL)as a crosslinking agent.Then,the antibacterial properties and whitening effect were evaluated.This research may provide a new approach to the research and development of natural,safe and efficient cosmeceuticals,and provide a theoretical reference for efficient application of BLF,TP and other natural products in functional cosmetics.The main research contents and results are as follows:(1)The nothing loaded TP/?-PL nanoparticles were prepared by ionic cross-linking method,using turnip TP and ?-PL as materials.The preparation conditions were optimized by using variable-controlling approach and taking the particle size and PDI dispersion index as the standard.The results showed that when TP concentration was 2-4mg/mL,?-PL concentration is 1-2mg/mL,TP/?-PL mass ratio was 2:1,The particle diameter was about 292.5±2.17nm,PDI dispersion index was 0.169±0.02,the shape is spherical and regular,the particle size distribution is concentrated and of good dispersibility.Thus,it could be used as a good nanocarriers loaded with BLF.(2)The preparation method of BLF/TP/?-PL nanoparticles loaded with BLF were optimized by single factor experiment with particle size distribution and embedding rate of BLF as index.The results showed that when the ratio of TP and ?-PL was 5:1,ratio of wall material and core material was 15:1,concentration of wall material was 3mg/mL,and the speed of constant flow pump was 200 mL/h,the particle diameter reached to 500?550nm,PDI dispersion index was 0.224±0.01,the shape is regular and uniform,and the embedding rate is 45.27%.The repeatability test showed that RSD<2.0%,the preparation process was feasible,repeatable and simple,thus,the expected result was obtained.(3)Using mouse B16 melanoma cell model to evaluate the whitening efficacy of BLF/TP/s-PL nanoparticles.After B16 melanoma cells were treated with BLF/TP/?-PL nanoparticles,the cell proliferation rate,tyrosinase activity and melanin content were measured respectively,and the effects were compared with that of arbutin.Results showed that the tyrosinase activity,the synthesis of melanin and proliferation rate of B16 cells were all negatively related to the drugs concentration.When the concentration is 100 mg/L,the inhibition rate reached at 69.68%,62.35%and 59.14%respectively,showing significant difference as compared to arbutin(p<0.05),which indicating an effective whitening effect of BLF/TP/?-PL nanoparticles.At the same time,IC50 safety evaluation showed that the IC50 of cell proliferation was greater than that of melanin production,which means a good security of BLF/TP/s-PL nanoparticles.(4)The antibacterial activities of BLF/TP/?-PL nanoparticles on gram-negative bacteria Escherichia coli and gram positive bacteria Bacillus subtilis were measured by Oxford cup assay method,then compared with that of ?-PL.The results showed that BLF/TP/S-PL nanoparticles had good antimicrobial effect on both kinds of bacteria and presented minimum inhibitory concentrations(MIC)of less than 0.625 mg/mL,the inhibitory effect was positively related to the concentration,slightly weaker than ?-PL at the same concentration.To sum up,BLF/TP/?-PL nanoparticles were composed of natural ingredients,prepared by a simple method,and itself had stable and homogeneous properties,strong antibacterial effect,whitening efficacy as well as safety,thus,it has a promising application prospects in the field of natural cosmeceuticals.
