Electrospinning,as a new technique for the preparation of nanomaterials,due to its simple equipment and operation as well as applicable variety,has attracted widely attention in biomedical,optical electronic,catalytic materials,security,environmental engineering and many other fields.Especially in biomedical field,such as drug delivery,tissue engineering scaffold and wound healing.This thesis mainly includes two parts.First of all,based on the research of oral squamous carcinoma,PLA electrospun membrane with cisplatin loaded and PLA-PVP composite membrane with cisplatin and chloroquine loaded were prepared.Nanofiber prepared through electrospinning is harmless,and can be degraded in human body to release drug so as to achieve the treatment,which has gained a lot of attention recently.Oral squamous carcinoma is one of the most malignant maxillofacial tumors in China.Traditional treatment methods are mainly composed of surgical removal and chemotherapy.Except our group,there has no report about drug-loaded electrospinning membrane applied in the treatment of oral squamous carcinoma recently.The main contents are as follows:1.The scanning electron microscope was used to observe morphology of fibers prepared by different concentrations of PLA and PVP spinning solutions.Optimum spinning conditions have been disscussed.Biocompatibility and degradation rate of PLA and PVP membranes have been tested.2.PLA fiber membrane with cisplatin loaded was prepared,and drug-loaded membrane was used to test the destruction to oral squamous carcinoma KB cells.This kind of membrane drug-release system could control drug release slowly,and had more durable and better effect.In addition,PLA-PVP composite fiber membrane with cisplatin and chloroquine loaded was prepared.The destruction test confirmed that chloroquine can enhance the effect of cisplatin on oral squamous carcinoma KB cells.In addition,the researches for oligomers electrospinning are less than polymer.Annular CBT is a kind of high performance resin material,which has good compatibility for most materials and composite materials have been widely used in construction,transportation,food packaging and many other fields.One of the important applications is to prepare linear PBT through ring opening polymerization.PBT is a kind of important engineering plastic,which has good mechanical properties and corrosion resistance.CBT,as a resin material,has chemical resistance so it doesn't dissolve in most organic solvents.Therefore,we adopted the method of melt spinning to prepare CBT fiber,and to prepare PBT fiber through CBT ring opening polymerization under the condition of Butylchlorodihydroxytin as catalyst.It is a new kind of one-step method that combine physical spinning with chemical reaction.The main contents are as follows:1.Preparation of CBT fiber through melt spinning has been investigated.The best spinning condition to prepare PBT fiber has been confirmed by changing the dosage of catalyst,spinning voltage and receiving distance.2.The scanning electron microscope was used to observe morphology of fibers.Infrared spectrum,differential scanning calorimetry,1H NMR and thermogravimetric analysis have been conducted to confirm that PBT fiber can be obtained by melt spinning,which have improved the research of oligomers electrospinning,and laid a theoretical foundation for further application.
