The solid state fermentation processes and raffinose extraction conbined with solid state processes of cottonseed meal was investigated in this study.Firstly,two strains(Lactobacillus plantarum,Lactobacillus fermenti)with high effieieney of gossypol detoxification,two strains(Bacillus subtilis,Canidia Albicans)with high effieiency of protein degradation;and a strain(Saccharomyces cerevisiae)with high effieiency of protein content improvement were sereened out.Single strain mixed with two or three bacteria fermentate,finally the result mixed fermentation of Lactobacillus plantarum,Lactobacillus fermenti and Saccharomyces cerevisiae to produce cottonseed meal with high quality.The free gossypol decreased from 1274.7 mg/kg to 375 mg/kg with a detoxification rate of 70.5%,soluble protein from reached 7.4 g/100 g,and free amino acid was 68.13mg/g.Single factor experiment was used to confirm optimum conditions.Based on the results,the optimum conditions of cottonseed meal werecomfirmed by mixed Lactobacillus plantarum,Lactobacillus fermenti and Saccharomyces cerevisiae fermentation,respectively.The marked impact of four fermentation factors were confirmed as follows: the ratio of substrate to water,fermentate temperature,incubation time and wheat bran adding.The optimum fermentation conditions of detoxification were confirmed using Design-Expert.The results show that detoxification was up to 80.38% under the optimum fermentation conditions: initial moisture content of solid substrate 53%,time at 44 h,incubation temperature at 30?,15% wheat bran in substrate.When raffinose were extracted from cottonseed meal using solvent,followed by the microbial solid-state fermentation based on the residue of cottonseed meal,the content of gossypol was greatly reduced and the raffinose was harvested.After cottonseed meal was extracted with the solvent 75:25(v/v)ethanol/water containing 1.4 mol/L citric acid for 2 h,the leaching rate of raffinose reached 96.4%.The purity of raffinose reached 91.3% after refining.Solid-state fermentation were carried out to produce feedstuff by Lactobacillus plantarum,Lactobacillus fermenti and Saccharomyces cerevisiae based on the cottonseed meal residue extracted for raffinose.The free gossypol decreased from 1274.7 mg/kg to 118 mg/kg with a detoxification rate of 90.74%.Finally,in order to compare nutritional quality of fermented cottonseed meals of five different processes,various indexes such as detoxification,phytic acid content,raffinose content,crude protein content,soluble protein content,free animo acid content,reducing sugar content,total viable count of Lactobacillus,the molecular weight of protein,organic acid,neutral protease,amylase and cellulase were examined,the rusults showed best process was raffinose extraction combined with solid state fermentation.
