Metal-organic frameworks(MOFs)are a kind of porous materials with periodic multidimensional network formed by hybridization of metal ions and organic ligands through self-assembly process.MOFs has become a hot research for researchers in the catalysis,sensing,gas storage and drug loading and other aspects have shown excellent performance.Particularly in drug delivery,due to its large specific surface area,and good biocompatibility has been good materials as drug carriers.In this paper,three kinds of nano-MOFs drug carrier with good biocompatibility were prepared,and then the functionalization of the drug carrier has been realized by post-synthesis modification,which achieved the combination of nano-MOFs with the drug and the potential active tumor targeting.In this paper,nano-ZIF-90 with particle size less than 300 nm was prepared by ultrasonic-stirring method and optimized reaction conditions.The structure were determined by Fourier transform infrared spectroscopy(FT-IR)and X-ray powder diffraction(XRD)and the Scanning electron microscopy(SEM)was used to determine the size of the morphology.DOX was used to modify on the surface of ZIF-90 nanomaterials.The solid-state nuclear magnetic spectrum,FT-IR,thermogravimetry(TGA)were used to convert the amino groups in the ZIF-90 ligand and the antidumor drug doxorubicin(DOX).TGA was used to determine the DOX successful modification.5-fluorouracil(5-Fu)was loaded and released on the nano-drug carrier DOX@ZIF-90.The amount of 5-Fu was loaded nano-drug carrier reaching 0.572 g/g.Because of instability for ZIF-90 under the contion of faintly acid,the release of tumor tissue,DOX release of 0.45 g/g,5-Fu release rate of 95 % or more.This is the first time drug carrier based on MOFs to achieve combined loading of two antineoplastic agents.The microstructures and sizes of nano-PCN-223 and nano-MOF-808 were prepared by heating refluxing method and optimized reaction conditions.The morphology and size of the films were determined by FT-IR,XRD,TGA and UV-vis spectroscopy(UV-vis)were used to modify and release anti-tumor drugs on the PCN-223 and MOF-808 materials by conducted the study of 5-Fu.The results showed that FA@MOF-808 loaded with 5-Fu up to 0.654 g/g,the release rate could reach 80.14 %;FA@MOF-808 material loaded 5-Fu was up to 0.846 g/g,the release rate was 78.13 %.The drug carrier is the first time to achieve the active active tumor targeting drug carrier of MOFs by coordination.
