Bellamya aeruginosa being as a potential ecotoxicological model organism,in this thesis,its laboratory conditions were optimized and screened firstly under artificial conditions using the growth as the endpoint.The results showed that the body could grow better with the following conditions: feeding fresh chlorella,culturing with the density of 6·L-1,keeping the water depth of 10-15 cm,using the soil as the sediment and the calcium ion being 30 mg·L-1.And MgCL2·2H2O could be used as the primary choice for anesthetics to snail.To accurate the stability of the reference gene,eight candidate reference genes expression levels in tissues were quantified in river snails exposed to different stresses for 10 days.The results showed that,ribosomal protein L7(RPL7)was the most stable reference gene in all samples tested.It was concluded that RPL 7 was a suitable reference gene in river snail for the quantification of gene expression under various environmental stresses.Finally,we have an initial research on Taihu Lake Basin Bellamya aeruginosa's sex ratio in Taihu Lake Basin.The results showed a high aberration rate and the bifurcated or webbed distortion of right or both tentacles occurred in approximately 2.7%.In distortion and normal tissue studies of the adult snails,we found the muscle tissue contain a lot of parasites embryo,its gastropods were smooth muscle in the normal tissues,and the testis which belong to the evolutionary type II sperm were containing a large number of mature cluster sperm.This knowledge will further be helpful for rearing and maintaining Bellamya aeruginosa as a promising model organism.
