To explore the feasibility of silk fibroin membrane functioned as a guide bone regeneration membrane(GBR barrier membrane).In this study,two methods were conducted to prepare silk fibroin membranes followed that the physicochemical properties as well as biological behaviors of silk fibroin membranes were investigated respectively.1)The papermaking process was used to prepare silk fibroin membrane,according to which a series of fibroin-collagen blended membranes were obtained at different mixing ratio.The physicochemical properties of the blended membranes were characterized.According to the SEM images,the membranes showed a complex network structure of silk fibers.The FTIR analysis showed the conformation of membrane is ?-sheets structure.In vitro,MC3T3-E1 cells and STO cells were employed to evaluate the cell compatibility of blended membrane.The result showed that cells seeded on the surface of the membranes could attach and stretch well.The in vivo experiments were performed by subcutaneously implanting blended membranes on the back sides of Sprague Dawley rats(SD rats).The rats were sacrificed at 1,4 or 8 weeks after surgery and evaluated by histology.Hematoxylin-eosin staining confirmed that inflammatory cell infiltration occurred in the pre-implantation near the membrane material;however,the inflammatory phenomenon slowed down over time.The SF2 group owned the similar biocompatibility to the commercial collagen membrane.Incubated 9 weeks,the membrane material maintained relatively complete.2)Lyophilized and dense silk fibroin membrane was fabricated and its mechanical behavior as well as biodegradability was investigated.The osteoconductive potency of the silk fibroin membranes was evaluated in a defect rabbit calvarial model.Silk fibroin membrane showed the modulated biodegradable and mechanical properties via ethanol treatment with different concentration.The membrane could prevent soft tissue invasion from normal tissue healing,and the amounts of new bone and defect closure with silk fibroin membrane were similar to those of commercially available collagen membrane.
