Korean soybean paste as a traditional food of Yanbian,not only delicious,but also nutrient-rich,which also contains a variety of functional ingredients that conducive to human health,including antioxidant,lowering blood pressure,anti-aging,inhibition of tumor growth and many other physiological functions.At present,there are many studies on the fermented products of soybean and soybean products,but the research on the quality of Korean soybean paste and the effect of inhibitory effects of Korean soybean paste on different tumor cells are still blank.Therefore,the results of this study can provide theoretical guidance for the fermentation process and basic research of Korean soybean paste,and promote the development of Korean food to the direction of functional food,and provide new ideas for cancer diet therapy.Objective1.Compare the quality of Korean soybean paste with different strains and the effect of different strains on the content of isoflavones in Korean soybean paste and determine the best fermentation scheme.2.Inoculation of different strains of Korean soybean paste in the determination of cancer components.3.To determine the inhibitory effect of Korean soybean paste on different tumor cells by fermentation according to the best fermentation protocol.Methods1.The effects of the fermentation time and the inoculated strains on the main components in the miso were discussed by single factor experiment,and then the optimal fermentation scheme of Korean soybean paste was determined and the quality was compared.2.Select the best fermentation program for miso fermentation,and then assaying the isoflavones of fermentative soybean paste.3.Select the fermentative Korean soybean paste which inoculation of Bacillus licheniformis and Aspergillus oryzae.Choose the Korean soybean paste conduct an in vivo test which fermentate of 15 days,30 days,45 days,60 days,75 days,90 days.Use the Korean soybean paste which fermentate for 90 days to prepare into different concentrations of soybean paste solution,the tumor cell inhibition rate was measured and conduct to an in vitro test.Result1.The contents of isoflavones were the highest at 90 days of the Korean soybean paste which were inoculated with three different Bacterias,which were 0.202%,0.196%and 0.230%respectively,there is the highest content of isoflavones which inoculated with Bacillus licheniformis and Aspergillus oryzae.2.The Korean soybean paste which inoculated with Bacillus licheniformis and Aspergillus oryzae in the same time,comprehensive evaluation of the highest score in the color,taste,flavour,and the highest content of isoflavones was the 90 days of fermentation.So Bacillus licheniformis and Aspergillus oryzae were selected for inoculation,fermentate for 90 days as the optimum fermentation conditions.3.In vitro test:Korean soybean paste had no toxic on human normal cells,and the inhibitory effect on the human kidney embryo Hek-293 cells was not obvious.The inhibitory rate of isoflavones was about 20%,while that of human cervical cancer Hela cells,human colon cancer HCT-116 cells and human hepatoma HepG2 cells have a significant inhibitory effect,the inhibition rate of them more than 70%,and has the best inhibitory effect on liver cancer HepG2 cells.In vivo test:the average tumor weight of the mice treated with the different fermentation days Korean soybean paste extractive was lower than that of the blank control group.With the increase of the fermentation time,the average tumor weight decreased gradually and the tumor inhibition rate increased gradually.The Korean soybean paste extract which fermentate for 90 days had the best inhibitory effect on the tumor,the tumor weight was 2.19 ± 0.61 g,the inhibition rate was 45.0%.The results indicating that the Korean soybean paste which fermented for 90 days can significantly inhibit the tumor growth in S-180 sarcoma transplantation Balb/c mice.ConclusionsKorean soybean paste obtained the best quality which inoculated Bacillus licheniformis and Aspergillus oryzae,and it obtained the highest content of isoflavones,and it had good inhibitory effect on tumor cells.
