Heilongjiang Province is an important food base for planting non-GM soybeans in China.There are excellent texture,high fat,high protein,and high isoflavones in soybeans,which play an important role in the domestic market and soybean product processing.However,with the fierce competition in the market,some problems of brand soybeans occurred in Heilongjiang province such as quality and security,and the authenticity of the brand,which played an important role in the research on the origin traceability.This research can find the characteristic index that characterize the origin of soybean,and it is also one of the effective way to achieve the protection of the quality of brand soybeans in our province and the discriminant analysis of the origin traceability.The isoflavone monomer content was detected with high performance liquid chromatography in different areas and soybean varieties in Beian Administration Bureau of Heilongjiang Farms and Land Reclamation Administration,and Nenjiang Northern Agricultural Development Co.Ltd of COFCO of Heilongjiang province in 2015 and 2016.Analysis of chemometrics methods such as variance,principal components,clustering,and discrimination were used to select the characteristics of isoflavones monomer.The feasibility for the origin of soybeans were determined with the characteristics of isoflavones monomer,the effects of year,origin,variety and their interactions on the characteristics of soybean isoflavones were also examined.Combining chemometrics methods the traceable indicators of effective isoflavones were screened out,and a discriminating model of origin was established and verified.To achieve the purpose of tracing the origin of soybean the characteristics of isoflavones monomer in soybean was stored in the database,and a traceability database of soybean isoflavones was established.Results were listed as following:1.Improved chromatographic conditions:The mobile phase was 0.1%acetic acid-water solution and 0.1%acetic acid-acetonitrile solution,gradient elution.The column temperature was 30°C,detection wavelength was 260 nm,the flow rate was 1.0 mL/min.The linear range of the six soybean isoflavone monomers was 0.25?g/mL to 10.00?g/mL,the determination coefficient of R~2?0.9993.The recoveries ranged from 99.62%to 114.0%,and the relative standard deviation RSD ranged from 0.96%to 7.99%.The results provided technical support for the detection of large quantities of soybean isoflavone monomer content in post-production traceability researches.2.Six kinds of soybean isoflavone monomer content in 51 soybean samples from different areas in Heilongjiang province in 2015 were detected,chemometric methods such as variance,principal components,clustering,and discrimination were used to screen the characteristics indexes of isoflavones monomer.The overall correct discrimination rate of the soybean production area is 84.3%,which has good effect and application value for the discrimination of soybean production area.3.Six kinds of soybean isoflavone monomer content were detected in 76 soybean samples from different areas in 2016 in Heilongjiang Province.Combined with chemometric analysis,the correct discrimination rate of soybean production areas and the correct identification rate of cross-validation reached 98.7%.It was proved that the origin tracing with characteristics of soybean isoflavones monomer was feasible and effective.4.The effect of origin,variety,year,and interactions between various factors on the content of soybean isoflavone monomers was investigated,and the characteristics of soybean isoflavone monomers directly related to the origin was screened.A discriminant model by chemometric analysis method was established to verify the discriminant analysis.With the selected soybean flavonoids,glycitin,genistin and other three kinds of soybean isoflavone monomer characteristics,the correct identification rate of soybean production areas reached 81.1%,which met the requirements of the discriminant analysis of origin.5.The soybean origin traceability database was constructed and verified by querying the soybean isoflavone monomer characteristic information.The results showed that the database can effectively distinguish the origin,year and variety of soybean by the characteristic index of soybean isoflavone monomer,which complete provided a reference for the research of soybean production area.In summary,using the improved HPLC technology to detect the content of soybean isoflavone,combined with the chemometrics method,a discriminant model and database was established to trace the origin of soybean origin in Heilongjiang province.This research provides a technical support for the further study of soybean production areas,variety identification technology,traceability technology and the construction of recall system for the use of soybean secondary metabolites as indicators.
