Bioactive glasses?BGs?have great application prospect in tissue engineering scaffold material,orthopedic,dental and drug carrier.BG nanofibers exhibit some novel physical and chemical properties due to their large surface area and size effect,and get more and more attention from researchers.Electrospinning is a popular technique for producing nanofibres in mesh or fiber mat morphology.However,the process is complex,high cost and low production rate,and the fiber diameter ranges from tens to hundreds nanometers and the thickness of the as-prepared scaffolds is in microns.Thus,finding an alternative method which is simple,low cost,environment friendly and high yield is of theoretical and practical significance.The aim of this study was to synthesize SiO2,60S40 C,58S and 58S/Gel nanofibrous scaffolds with 3-D network,using BC nanofibers as the template and evaluate the in vitro bioactivity and biocompatibility.Amorphous SiO2 and 60S40 C nanofiberous scaffolds with 3-D network structure were synthesized by using BC and preliminary calcified BC as template.The average diameter of SiO2 and 60S40 C nanofibers was 18 nm and 25 nm,with roughness of 3.56 nm and 3.42 nm,respectively.Mercury intrusion porosimetry?MIP?and nitrogen adsorption results showed that SiO2 and 60S40 C have suitable pore structure for cell growth.The in vitro mineralization results showed that 60S40 C nanofibrous scaffold has high bioactivity.The biocompatibility of SiO2 and 60S40 C nanofibrous scaffolds were evaluated by fluorescence staining,CCK-8 inspection and ALP activity evaluation using the rat osteoblast.The results showed that 60S40 C nanofibrous scaffold has better biocompatibility in vitro.On the basis of the preparation of SiO2 and 60S40 C nanofibrous scaffold,58 S,58S/Gel-1,58S/Gel-2 and 58S/Gel-4 nanofibrous scaffold were prepared.The average diameter of 58 S and three kinds of 58S/Gel nanofibers was 31 nm,45 nm,60 nm and 76 nm,with surface roughness of 3.25 nm,1.54 nm,1.42 nm and 1.28 nm,respectively.MIP and nitrogen adsorption results showed that 58 S and 58S/Gel have suitable pore structure for cell growth.The in vitro mineralization results showed that 58 S and 58S/Gel nanofibrous scaffold have high bioactivity.The biocompatibility of 58 S and 58S/Gel nanofibrous scaffolds were evaluated by fluorescence staining,CCK-8 inspection and ALP activity evaluation using the rat osteoblast.The results showed that 58 S and 58S/Gel nanofibrous scaffolds could promote osteoblast proliferation and differentiation.58 S and Gel have synergistic effect on the proliferation and differentiation of osteoblast.Among them,58S/Gel-2 nanofibrous scaffold promoted osteoblast proliferation and differentiation most significantly and has the potential application in the field of bone tissue engineering.
