Nowadays,nanotechnology has increasingly been used for the design of drug delivery,photodynamic therapy and imaging modalities in order to further improve efficacy in cancer treatment.However,due to the complexity and diversity of tumor diseases,single functional nanomaterials,such as:only used as delivery carrier or photodynamic therapy,cannot achieve effective treatment:Therefore,the current thesis work aims to develop a new multifunctional nanomaterial for the high efficiency treatment of cancer.Mesoporous silica nanoparticles(MSNs)have inspired intensive research interest due to their excellent features such as enhanced loading capacity,large surface area,rich surface chemical properties,and their potential applications in nanomedicine.In this thesis,a multifunctional nano-drug based on MSNs was fabricated for cancer treatment and anti-bacteria.Monodisperse MSNs were synthesized via a modified micro-emulsion method.By embedding carbon dots and a photosensitizer(Rose Bengal)in core/shell MSNs respectively,a multifunctional nano-drug was obtained,and the high efficacy of the nano-drug for the inhibition of tumor in vitro and antibacteria was demonstrated.The main results are summarized below:(1)The core/shell MSNs were synthesized via a modified micro-emulsion method.The relationships between the morphology of the MSNs and the reaction time,temperature and the amount of silica source were fully studied.Finally,the core/shell MSNs with size of 104 ± 6.0 nm were obtained,the pore size of the MSNs were 3 nm,and the MSNs were high uniform nanoparticles with good dispensability in ethanol.(2)The multifunctional nano-drug was obtained by embedding carbon dots and Rose Bengal in MSNs during the two steps process of synthesis,respectively.The optical property,the production of 1O2,drug loading capacity and loading efficiency were fully characterized.(3)Tumor therapy and anti-bacteria research of the multifunctional nano-drug.Firstly,the capability of imaging-guided drug delivery of the multifunctional nano-drug was studied by confocal microscopy images;then the high efficacy of the multifunctional nano-drug for the inhibition of tumor growth in vitro was fully demonstrated,more than 90%cancer cells(H1299)were killed by this nano-drug at the concentration of 100?g/mL.Finally,the anti-bacterial effect of this nano-drug was fully studied,the bacterias were effectively inhibited by this nano-drug under the concentration of 100 ?g/mL.The above revealed great potential of this nano-drug as noninvasive medicine for cancer therapy and bacterial inhibition.
